Objective:The genetic polymorphism of the 5 Y-chromosomal Short Tandem Repeats (STR) locis in Chinese Korean and Han population in Yanbian area was detected and analyzed to observe the suitability of this system in the forensic medicine studies of individual discrimination, paternity identification, and human genetics.Methods:DYS460、DYS462、DYS463、DYS635、Y-GATA A10 five locis of 200 unrelated Korean and 200 unrelated Han individuals were tested by PCR, polyacrylamide gel electrophoresis, and silver staining. All the data were analyzed with Arlequin3.11 and Excel software.Results:There are 5 and 4 alleles found by DYS460 loci in the Han Chinese and Korean population, which are No.8 to 12 alleles. There are 3 alleles found by DYS462 loci in the Han Chinese and Korean population, which are No.11 to 13 alleles. There are 6 alleles found by DYS463 loci in the Han Chinese and Korean population, which are No.17 to 23 alleles. There are 9 and 7 alleles found by DYS635 loci in the Han Chinese and Korean population, which are No.7 to 25 alleles. There are 6 and 5 alleles found by Y-GATA A10 loci in the Han Chinese and Korean population, which are is No.11 and 13 to 17 alleles. The gene diversity (GD) of 5 locis in Han population are 0.6787、0.3579、0.6677、0.7708, and 0.7071, while the GD of 5 locis in Korean population are 0.6600、0.4449、0.6270、0.7212, and 0.7013. Additionally,140 and 118 haplotypes are detected from Han population and Korean population by accumulation of 5 Y-STRs, their haplotype diversity (HD) are determined to be 0.9935 and 0.9902.Conclusion:DYS462 genetic polymorphism is lower in the 5 locis, while other locis display good gene polymorphism and highly applied value. The Fst and P-value show 5 Y-STR locis exhibit obvious differences in different areas and ethnic. So, it is confirmed that 5 locis’gene polymorphismi2s suitable for forensic medicine studies of individual discrimination, paternity identification, and human genetics.
