Objective:By investigating the genetic polymorphisms of five Y-STR locis inthe Korean and Han populations in Yanbian,Jilin Province,statistical analysis of gene and genotype frequency distributions and related genetic parameters of five Y chromosome STR locis were obtained,providing basic data for forensic applications;Combined with the 48 Y-STR locis obtained in the previous study of this laboratory,the differences among different groups were analyzed to determine the genetic characteristics of Y-STR in Korean populations;A genetic locus rich in genetic data was selected to analyze the genetic distance between 23 ethnic groups in China and between foreign populations.A multidimensional scale analysis scatter plot was made based on the genetic distance to compare ethnic and racial differences.Methods:Five single-copy Y-STRs(DYS549,DYS576,DYS578,DYS588,DYS643)of 200 Yanbian Korean and 200 Han unrelated male individuals weretested by PCR and detected by polyacrylamide gel electrophoresis with silver staining techniques.The results were analyzed with Excel 2017,SPSS and GenAlEx 6.503.Results:There were 5?8?5?9?7 alleles detected from Yanbian Korean population in single-copy locus DYS549?DYS576?DYS578?DYS588?DYS643,and gene diversity were 0.5884?0.7625?0.5575?0.3434?0.7494 while there were 5?8?6?7?7 in Han population and gene diversity were 0.5801?0.7966?0.6724?0.3523?0.6981.In total,200 unrelated individuals were analysed and 115 haplotypes were observed and reached haplotype diversity(HD)of 0.9874 in Yanbian Korean groups;in total,200 unrelated individuals were analysed and 130 haplotypes were observed and reached haplotype diversity(HD)of 0.9932 in Yanbian Han population.In our laboratory,we analyzed 53 Y-STR locis in Yanbian Korean and Han male individuals,and 282 and 292 alleles were detected respectively in 48 single-copy Y-STR locis,and 145 and 146 haplotypes were detected respectively in multiple copy locis(DYS385a/b,DYS459a/b and DYS464);The GD values of 53 Y-STR locis ranged from 0.0199 to 0.9790 in the Yanbian Korean population,and the GD values ranged from 0.0775 to 0.9818 in the Yanbian Han population;53 Y-STR locis were accumulatived and 200 haplotypes were found respectively inYanbian Korean and Han populations,and the haplotype diversity(HD)is 0.995;The genetic distance results show that the genetic similarities between the same ethnic groups in different regions are not obvious,different ethnic groups have significant genetic differences even in the same geographic area;Multidimensional scaling analysis(MDS)scatter plots further confirmed the correlation between population differences and ethnic origins and geographic locations.Conclusion:The five Y chromosome STR locis screened in this study enriched the genetic data of Yanbian Korean population in Jilin Province.The genetic data of 53 Y-STR locis in the Yanbian Korean ethnic group obtained by this laboratory accurately reflect the genetic information characteristics of the Yanbian Korean population,providing important support and guidance for the application in forensic practice,and providing important reference data for forensic testing,population genetics and ethnology.Through comparison with domestic and foreign populations,the existence of genetic polymorphism differences between different populations was further confirmed,and geographical distances between groups will largely influence this difference.
