Study On The Mechanism Of Fujiu Tie Ointment On Airway Neurogenic Inflammation In Mice With Bronchial Asthma

Purpose:To observe the effects and therapeutic effects of Fujiu patch on Substance P(SP),neurokinin 1 receptor(NK-1R)and immediate early gene(c-Fos)in the lung tissue of mice with bronchial asthma,and to explore the mechanism of action,so as to provide experimental basis for clinical promotion of Fujiu patch in the treatment of bronchial asthma.Materials and methods: 50 BALB/c magnetic mice were selected and randomly divided into normal control group(normal group),bronchial asthma model group(model group),western medicine positive control group(dexamethasone group),fujiu plaster group,and false acupoint plaster group(Vaseline patch group),with 10 mice in each group.Ovalbumin(OVA)was used to induce the mouse model of bronchial asthma.At the same time,except for the normal group and the model group,the other groups were given corre SPonding intervention treatment.The general state of mice was recorded during the experiment.At the end of the experiment,pathological changes in the lung tissues of mice were observed by HE staining.Immunohistochemical(HIC)method was used to observe the expression of SP protein in lung tissue of mice in each group.Western Blot was used to determine the expression of NK-1R and c-Fos protein in lung tissues of mice in each group.Results:1.General state of mice: compared with the normal group,the behavior of mice in the model group was as follows: reduced food intake and water intake,bristle and hair stood silent,nodding and breathing occurred when stimulated by atomization,deepened and accelerated breathing,tiredness,instability in standing,ininence and other typical asthmatic attacks;Compared with the model group,the hair of the dexamethasone group and the fujiu patch group were slightly darker,and the asthma like attack was lighter than that of the model group.The status of the vaseline patch group was slightly better than that of the model group,but the difference was not significant.Compared with the dexamethasone group,the mental state,mobility,feeding,and water intake of the mice in the Vjiu patch group were better.2.Pathological changes of lung tissue: Compared with the normal group,the pathological sections of lung tissue in the model group showed a large number of inflammatory cell infiltration,bronchial wall thickening,even red mucus plugs,disorderly arrangement and detachment of epithelial cells in bronchial mucosa,and thickening of alveolar septum,etc.Compared with model group,the degree of inflammatory cell infiltration in lung tissue of mice in dexamethasone group and Fujiu paste group was reduced to different degrees,and the degree of inflammation in lung tissue of mice in Vaseline patch group was not significantly different from that in model group.Compared with the dexamethasone group,the bronchial wall of the lung tissue of mice in the Fujiu patch group was thicker and the infiltration of inflammatory cells was more serious.3.The results of detecting SP protein expression in mouse lung tissue by immunohistochemistry showed that the positive expression of substance P in mouse lung tissue was brown-yellow region,which was mainly concentrated in the cytoplasm.Compared with normal group,the positive expression area of substance P in lung tissue of model group was significantly higher than that of normal group(P < 0.01).Compared with the model group,the positive expression area of substance P in lung tissue of dexamethasone group and Fjiu patch group was significantly less(P < 0.01),and the positive expression area of substance P in lung tissue of Vaseline patch group was less than that of the model group,but the difference was not significant(P BBB 0 0.05).Compared with the dexamethasone group,the positive expression area of substance P in lung tissue was larger in the Fujiu patch group,but the difference was not significant(P > 0.05).4.Western Blot analysis of NK-1R and c-Fos in lung tissue of mice showed that the relative protein expression levels of NK-1R and c-Fos in lung tissue of mice in other groups were significantly higher than those in normal group(P < 0.01);Compared with model group,the relative expression levels of NK-1R and c-Fos protein in lung tissue of dexamethasone group and Fujiu patch group were significantly decreased(P < 0.01),while the relative expression levels of NK-1R and c-Fos protein in lung tissue of Vaseline-patch group were not significantly decreased(P > 0.05).Compared with the dexamethasone group,the relative expression levels of NK-1R and c-Fos protein in lung tissues of the Fujiu plaster group were slightly higher,but there was no statistical difference(P > 0.05).Conclusion:1.BALB/c mice with bronchial asthma can be successfully established by induction of BALB/c magnetic mice with OVA.2.Fujiu paste and dexamethasone can down-regulate the expression of SP,NK-1R and c-Fos,and dexamethasone has a more obvious down-regulation effect.3.Fujiu patch and dexamethasone can reduce airway inflammation in mice with bronchial asthma,and dexamethasone is more effective.But in improving the general state of mice,the effect of Fujiu patch is more obvious.
