The Effects Of Chinese Wenshenbuxu Decoction On Liver CHOL/LDL/TSHR In Hypothyroidism Rat

Aim:This study was to perform the effect of Wenshenbuxu decoction on blood lipids,liver enzymes,thyroid function and thyroid stimulating hormone rece ptor(TSHR)expression in the liver of hypothyroidism,provide theoretical support for Chinese medicine intervention abnormal lipid metabolism caused by hypot hyroidism.Methods:Male SD rats were randomly divided into blank group (BG),mo del group, model group was gave 0.05% propylthiouracil (PTU) distilled water. After the modeling,the model group were randomly divided into model group (PTU),the Western Medicine group (L-T4),Wenshenbuxu low dose group (LDG), Wenshenbuxu middle dose group (MDG),Wenshenbuxu high dose group (HDG), administered for 4 weeks different drugs.During the experiment,recorded food i ntake,monitored body weight and anus temperature.Four weeks after administrati on,took blood from femoral arterym to exam cholesterol,LDL,liver enzymes,thyr oid function;Liver histology were observed using HE staining;TSHR expression on liver were detected by Immunohistochemical assay.Result:Compared with the blank group:The model rats eat less,the body w eight grow slowly,anus temperature decreased(P<0.01);serum CHOL,LDL,ALT,T SH increased(P<0.01),AST did not change significantly(P>0.05),cycling in T3, T4 decreased(P<0.01);the expression of liver TSHR showed a rising trend (P =0.063);thyroid HE staining showed hypothyroidism change,liver HE staining s howed express normal. After treatment,compared with the model group:the HD G and L-T4 group rats significantly recovery diet (respectively,P<0.01,P<0.0 5);HDG significantly increased body weight(P<0.01);the MDG and the HDG a nus temperature rises significantly (respectively,P<0.05,P<0.01);the high dose group CHOL,LDL decreased significantly (respectively,P<0.05, P<D.01);Wensh enbuxu decoction of each dose group and L-T4 group ALT,TSH was significan tly decreased (P<0.01);the HDG, MDG, L-T4 group T3,T4 increased significan tly (all P<0.01),low dose group T3 significantly increased (P<0.05);High dose group TSHR also have decreased trend.Conclusion:Hypothyroidism can cause blood lipids elevated.Wenshenbuxu decoction in order to alleviate the general situation of hypothyroid rats,partial r estoration of TSH,T3,T4 levels,reduce cycle CHOL,LDLcontent,there is a tende ncy down TSHR expression.Which can be used as a research direction that red uce blood lipids.
