Effect Of Wenshenbuxu Decoction On The Treatment Of Hypothyroidism Rats And The Expression Of TR And TSHR In Rats

Objective: To observe the therapeutic effect of Wenshenbuxu decoction on hypothyroidism model rats,and to explore its mechanism.To provide theoretical and experimental basis for the clinical treatment of Wenshenbuxu decoction.Methods: Male SD rats were randomly divided into blank group group,model group,and model group provided 0.05 % Propylthiouracil(ptu)distilled water model.After the success of the model,the model group randomly divided into western medicine group,low dose group,Chinese traditional Chinese medicine dose group,high dose group of traditional Chinese medicine.After 4 weeks of treatment,the general situation of rats was observed during the experiment,the diet was recorded,the body weight and body weight were monitored.At the end of the experiment,blood was taken from femoral artery and removed the pituitary,thyroid and liver to be tested.Thyroid function(TSH,T3,T4,FT3,ft4),liver function(ALT,AST),blood lipid(LDL,LDL),renal function(SCR,bun)were measured by ELISA.The expression of thyroid hormone receptor(tr)and thyrotropin receptor(Receptor)in pituitary,thyroid and liver were detected by immunohistochemistry.Observe pituitary,thyroid and liver histology by HE method.Results: 1.The symptoms of spleen deficiency syndrome appeared in model group,the level of T3,T4,FT3,ft4 decreased(p < 0.1)and TSH(p < 0.0),and the pathological changes of thyroid gland.2.The traditional Chinese medicine and western medicine group can significantly increase the level of T3,T4,FT3,FT4(p <0.01),reduce TSH concentration(p < 0.01).The western medicine group improved the thyroid function is better than traditional Chinese medicine.But the traditional Chinese medicine group is better than the western medicine group on relieving of symptoms.The ALT,CHOL and LDL of the model group increased significantly than the blank group(p < 0.01);There was no significant change in AST(p > 0.05).The western medicine group and Chinese medicine group can reduce the level of ALT,CHOL and LDL(p < 0.01),the Chinese medicine group was better than the western medicine group in improving the level of CHOL and LDL(p < 0.01).4.Compared with the blank group,the BUN and SCR of model group increased significantly(p <0.01).The western and Chinese medicine group can reduce the level of BUN and SCR(p < 0.01).The western medicine group and traditional Chinese medicine dose group is the best.5.Compared with the blank group,the expression level of TR in the pituitary and thyroid in model group decreased significantly(p < 0.01).The expression of TR in liver was significantly increased(p < 0.01).Compared with the model group,the western medicine group and the Chinese herb group can increase the expression level of TR in pituitary and thyroid,and reduce the expression level of TR in the liver.In the expression of pituitary,thyroid and liver,the western medicine group is more obvious than the traditional Chinese medicine group.6.Compared with the blank group,the expression levels of TSHR in the pituitary,thyroid and liver of model group increased significantly(p < 0.01).Compared with the model group,the western medicine group can reduce the expression of TSHR in the pituitary,and not reduce the expression level of TSHR in thyroid and liver.The Chinese medicine can reduce the expression level of TSHR in pituitary,thyroid and liver.7.The pituitary,thyroid and kidney tissues of model group appeared pathological changes,the liver did not see pathological changes.After treatment,the western medicine group and traditional Chinese medicine can improve the pathological damage of pituitary,thyroid and kidney.Conclusion: 1.The rat model of spleen hypothyroidism was successfully established by using the method of free drinking Propylthiouracil suspension.2.Wenshenbuxu decoction is superior to western medicine in improving the general symptoms of hypothyroidism rats;Wenshenbuxu decoction can increase the level of T3,T4,FT3,FT4 and reduce the level of TSH.3 Wenshenbuxu decoction can improve the level of CHOL,LDL,ALT,BUN and SCR,and have the pathological injury effect of repairing target organ.Under physiological conditions,the expression levels of TR and TSHR in pituitary,thyroid and liver were: liver > pituitary > thyroid gland.Wenshenbuxu decoction can regulate the expression of TR and TSHR in pituitary,thyroid and liver.This may be the partial mechanism of the therapeutic effect of Wenshenbuxu decoction.5.No Wenshenbuxu decoction was found to have hepatotoxicity.
