Objective:Observe the change of renal function in SD rats with Hypothyroidism, investigate the correlation between Hypothyroidism and renal function damage,observe Wenshenbuxu Decoction influence on renal function in rats, and confirmed the therapeutic effect of Wenshenbuxu Decoction to renal function damage in SD rats with Hypothyroidism.Methods:60 rats in total, select 50 rats Randomly, Feed them the PTU drug solution (distilled water with 0.05% concentration) cause hypothyroidism rats, the way of free drinking water for 4 weeks.After building model rats, random grouping of rat:model group, western medicine group, low doses of Wenshenbuxu Decoction group, moderate doses of Wenshenbuxu Decoction group, high doses of Wenshenbuxu Decoction group, each 10 in each group. Each group respectively with corresponding drug lavage for 4 weeks, model group and normal group was given distilled water to fill the stomach. After the experiment,Through puncturing femoral artery collected blood,Detect TSH, T4, T3, FT4, FT3, SCr, BUN, CysC with enzyme linked immunosorbent (ELISA); Taking thyroid tissue and kidney tissue section stained by HE was observed for the morphological.Results:1.With PTU feeding rats after 8 weeks, TSH of model group rats was obviously higher than that of normal group in rats. Model group rats T4, T3, FT4, FT3 significantly lower than that of normal. Between the treatment groups, The western medicine group of TSH, T4, T3, FT4, FT3 best recovery, and TSH return to normal levels.2.With PTU feeding rats after 8 weeks, Model group rats CysC, SCr and BUN were significantly higher than those of normal. Between the treatment groups, moderate and high doses of Wenshenbuxu Decoction groups rats CysC and western medicine group rats Cys, no difference. Moderate doses of Wenshenbuxu Decoction group rats CysC reduced to normal levels.High doses of Wenshenbuxu Decoction group rats CysC is higher than normal. Moderate and low doses of Wenshenbuxu Decoction groups rats SCr levels were lower than western medicine groups'.and reached normal levels.SCr of moderate doses of Wenshenbuxu Decoction group rats was obviously lower than that of western medicine group'.Moderate and low doses of Wenshenbuxu Decoction groups rats BUN and western medicine group rats Cys, no difference.The moderate doses of Wenshenbuxu Decoction group BUN reached normal levels in rats.Rats BUN level of low doses of Wenshenbuxu Decoction group was obviously higher than that of normal group of rats.3.CysC, BUN and TSH levels are related. SCr and TSH, FT3 has no obvious correlation. CysC, BUN and T4, T3, FT4, FT3 level negative correlation.4.Model group rats for part of glomerular moderate atrophy, reduced volume, dramatically reduce glomerular number did not see.Moderate doses of Wenshenbuxu Decoction and western medicine groups in the rat kidney are characterized by normal kidneys.High and doses of Wenshenbuxu Decoction group rats as part of glomerular mild atrophy, reduced volume, dramatically reduce glomerular number did not see.Conclusion:1,0.05% PTU solution can create the model of hypothyroidism SD rats, Wenshenbuxu Decoction and L-T4 can treat hypothyroidism, but the effects of L-T4 on bone metabolism may be bader than the influence of Wenshenbuxu Decoction.2,Rat renal damage degree and degree of hypothyroidism were positively correlated.3,Building Hypothyroidism rats renal damage successfully, Wenshenbuxu Decoction and L-T4 can treat renal damage in rats.moderate doses of Wenshenbuxu Decoction group is The best therapeutic doses.4,This experiment doses in rats with renal toxicity...
