| Anemone obtusiloba is a grassland on the more common weeds,for livestock not eating.In this paper,11 RAPD genomic DNA markers were used to analyze the genetic diversity of 11 populations in four populations,and the genetic diversity was analyzed with the ecological factors of four regions.Experimental results show:The order of size was as follows: Maqu> Luqu> Azi> Hezuo;four plots of soil(P <0.05).The content of available phosphorus in soil was extremely high,and there was no significant difference among the three soil samples(P <0.05).The total phosphorus content was very high,(P <0.05).The content of available nitrogen was very high,but there was significant difference(P <0.05),and the order of the total phosphorus was higher than that of the control group(P <0.05)).There was a significant difference in the available K content between the soils of Azi,Maqu and Luqu(P <0.05).There was no difference between the Hezuo and the Arabian and Maqu,and the order of the size was as follows: Azi> Maqu> Luqu,Hezuo> Luqu.The optimal reaction system and amplification procedure of RAPD-PCR were determined by orthogonal test and single factor test: total system 20μL,DNA template 30 ng,primer 0.4m M / L,Taq DNA polymerase 0.5U,d NTPs 0.4 m M 5 min 30 sec,denaturation 94 ℃ 1 min 10 sec,annealing 37℃,1 min 40 sec extension 72 ℃ 1 min 10 sec,continued filling 72 ℃ 6 min 30 sec,cycle number 40.The highest percentage of polymorphic loci in the population of Maqu yellow(MQY)was the lowest,and the geographical loci were the lowest,and the polymorphic loci of white populations were lower than those of yellow and light group of.The percentage of total polymorphic loci is higher than that of sub-populations(except MQY).The order of gene diversity and the percentage of polymorphic loci in Shannon information index and Nei’s index estimation are basically the same.The genetic variation of A.obtusiloba mainly occurs within the population,but some genetic differentiation has occurred between sub-populations.The second branch is the yellow flower,the shallow and the white population,the Maqu yellow and the light-colored population;the second is the yellow flower,the third is Azi yellow,light-colored population and Maqu white population.The results showed that the lower the altitude,the lower the soil nutrient content,the higher the genetic diversity of the populations,the higher the genetic diversity,the soil water content,the available nitrogen(N)content may play a role in the geographical distribution of A.obtusiloba. |
