Research On A Kind Of Qufeng-zhitong Ointment

Qufeng-zhitong ointment,is a preparation developed from famous and experienced Monglian medicine, which is composed of fungi and plants(Fomes fomentarius, Inonotus obliquus, Ramulus Mori, Aralia chinensis, Gentiana macrophylla Pall, Paeoniae Radix Rubra). It has been used to treat joints diseases and shown significant effect on arthritis.To optimize matrix composition of traditional Qufeng-zhitong ointment. Response surface methodology(RSM) was employed to investigate effects of four factors including the ratio of oil phase to water phase and the amount of stearic acid, glycerin, 72 and 721 emulsifiers on matrix formulation, with appearance, ductility, viscosity, PH value and centrifugal stability as comprehensive evaluation index. The best proportion of Qufeng-zhitong matrix prescription was as following: glycerin 16.5 g, 72 and 721 emulsifiers 25.5 g, stearic acid 23.5g, the ratio of oil phase to water phase 1:7.8.Qufeng-zhitong oinment with good gloss, good moisturizing and easy to apply, non-greasy,improved the traditional white plaster formulation contaminated clothing, suffering from skin allergies and other shortcomings. The new formulation in room temperature six month for observation, the results of the indicators and stability ointment no significant changes, indicating excellent preparation, performance stable formulation. In acute toxicity test 2.5 times the human dose of high usage were used to rats, have not yet seen any abnormal reactions and deaths, contrast each administration with the saline group rats body weight was no significant.The results show Qufeng-zhitong ointmentss had no-toxic external use.To investigate the effect and mechanisms of Qufeng-zhitong ointments on adjuvant arthritis rats. Arthritis severity was evaluated by body weight loss, paw swelling, arthritic score, and histological changes.In addition, the serum levels TNF-α, IL-1, SOD, MDA, GSH, NO were detected using ELISA method. Another portion of blood was used for blood parameters analysis. Through cell culture,the effects of the medicine on AA rats splenic cells multiplication capacity were studied. The influence of celiac macrophage cell culture fluid of AA rats on mice thymine multiplication capacity under the medicine was evaluated. Expressions of MMP-10 and VEGF in cartilage were detected using immunohistochemical analysis.Qufeng-zhitong ointments could significantly ameliorate paw swelling and arthritis score, down-regulate the levels of MDA, IL-1, TNF-α, NO and up-regulate the SOD and GSH levels in serum. It inhibited the splenic cells multiplication capacity and influence of celiac macrophage cell culture fluid of AA rats on mice thymine multiplication capacity, also effectively inhibit joint hyperplasia.The results suggested Qufeng-zhitong ointments possesses substantial anti-arthritis and antioxidant activities.
