Malania oleifera as a unique single species of plants in China,is a unique ancient wood oil species in China.Garlic fruit only grows in a small amount in the western mountainous area of guangxi and the eastern region of yunnan.Although the content of neuronic acid in garlic seed kernel oil is still high,there are still a few differences in the content of each component.Nervonic acid have a variety of functions such as promoting the proliferation and differentiation of nerve cells,making dull brain cells active and thereby delaying brain aging,and regulating blood lipid and blood sugar.Researchers studying the genetics and metabolism of ultra-long fatty acids detected that gene expression and thus the metabolism of fat and energy in the body were affected by changes in the amount of neurotic acid that could be obtained from a daily diet.Neurological disorders such as insanity and attention deficit disorder are also linked to levels of neurotic acid in the body.Many physiological and pharmacological activities of Nervonic acid are valuable in the field of medicine.For example,to improve the intelligence of infants and young children,the treatment of alzheimer’s disease and physical immune deficiency disease,and so on.Reported nervonic acid detection methods,mainly by gas chromatography is given priority to,its high boiling point,because the nerve acid needed for complete gasification temperature is very high,so usually when testing the methyl or ethyl esterification,into the boiling point lower ester compounds,complex and processing steps in the chemical reaction process is easy to the condition of the material was not completely,determination value caused some problems such as error is bigger.The raw material used in this experiment was the seed kernel of garlic nut from oil tea research institute of guangnan county,wenshan autonomous prefecture,yunnan province.The extraction process of garlic nut kernel oil by ultrasonic assisted method,the saponification and acidification process of mixed fatty acids in garlic nut kernel oil and the separation and purification process of neuronic acid in garlic nut kernel oil were systematically studied.High performance liquid chromatography(HPLC)has the advantages of high detection power,simple steps and easy operation,and avoids the process of derivatization.In this study,a new method was established for the determination of neuronic acid concentration by high performance liquid chromatography(HPLC),and the optimal ultrasound-assisted extraction of garlic seed kernel oil was optimized by response surface method.The experimental contents and results are as follows:1、.The neuronic acid in the seed kernel oil of garlic fruit was extracted and purified.The content of neuronic acid in the seed kernel oil of garlic fruit was determined by high performance liquid chromatography(HPLC).Column temperature:26℃;Detection wavelength:205nm;We’re going to put in 10 seconds.HPLC method was optimized to determine the optimal proportion of the mobile phase of the neuronic acid content(methanol-acetonitrile-tetrahydrofuran-0.4%acetic acid solution was 12:8:5:5),and the peak time of the neuronal calculation sample was basically the same as that of the standard product at about 18min.2、Uses the ultrasonic assisted extraction technology,using petroleum ether solution to extract of garlic fruit seeds oil,the main factors of the extraction process of single factor experiment and solid-liquid ratio,extraction temperature,ultrasonic time and so on conditions on the fruit oil seeds extract of garlic,examined again through the response surface optimization experiment,the experiment of the optimal extraction technology for:30 min ultrasonic time,extraction temperature 60℃ material liquid than 1:14(g/ml).The theoretical oil yield is 40.534%,while the actual oil yield is 40.011%,which is basically consistent.
