Capparis Spinosa L., originated from dry regions in west or central Asia, belongs tothe plant family of Capparidaceae and Genus is Capparis. In China, it’s mainlydistributed throughout Xinjiang and Tibet, and is used as a folk medicine for the treatmentof rheumatoid arthritis and gout. In order to develop a new drug on anti-rheumatoidarthritis by using Capparis Spinosa as the raw material,we have done a series of study.Firstly, we determined the prescription of ointment matrix and the preparation process byexperimental design optimization. Secondly,Capparis Spinosa was used as the main drugof ointment and the adjuvant arthritis model was adopted to investigate thepharmacological effects of Capparis Spinosa ointment on rheumatoid arthritis treatment.Finally, the stability and the quality standard of the ointment were studied. The mainlyresults are presented as follow.(1) Based on the theorical study of matrix prescription, we had determined thecomponent of the matrix and excipients. Four different prescriptions were tested by us andthe best prescription was present as follow: Stearic acid10g, Setanol30g, Span-806g,BHT3g,5563g was used as oil phase and glycerol20g, Ssorbic acid1g, Azone10g,Triethanolamine1g, Tween-802g, water206g was used as water phase. We furtherstudied the optimum preparation process in the aspect of emulsification time andtemperature. The preparation process was heating the oil phrase(without556) and thewater phrase to85℃separately then slowing pouring the oil phrase into the water phrasemixing them evenly in5mins adding the556when temperature fail to room temperatureto get the matrix and using petroleum ether abstract, ethyl acetate abstract and the watersoluble extract of the Capparis Spinosa L. ethanol extract as main drug. The matrix andmain drug were made into three kinds of Capparis Spinosa ointment samples in certain ration to evaluating the effect of rheumatoid arthritis treatment.(2) The study of pharmacological effects and mechanism of Capparis Spinosaointment on rheumatoid arthritis: We investigated the pharmacological effects of threedifferent kinds of Capparis Spinosa ointment by using the AA rat as the model, and thesample3which the main drug is the remaining of the Capparis Spinosa L. ethanol extractcan dramatically ameliorate the weight lose, footpad swelling and fall of IL-1β,TNF-αlevel caused by AA. However, all of the three samples did not influence the immuneorgans remarkably. Preliminary results show that reducing the serum level of IL-1β andTNF-α might be the major mechanism.(3) The study of the stability and the quality standard of Capparis Spinosa ointment:We mainly investigated the physicochemical property, stability and stimulating of thesample3. The ointment is relatively stable that it can maintain its property during a longtime in room temperature and has no stimulate to skin. According to the pharmacopeia weset the amount of flavones of the ointment no less than2%as a major quality standard.The work above laid the foundation for the development of a new drug onanti-rheumatoid arthritis, which using Capparis Spinosa extract as the main drug.
