Objective:Buzhong Yiqi Decoction (BZYQD),which is "Yiqi Shengyang" On behalf of the formula, act from" Treatise on the spleen and stomach" by Li Dongyuan, in the clinical treatment of diarrhea spleen deficiency frequently. This experiment study on histopathology and nutrient absorption of angle, modifird diarrhea spleen deficiency animal and investigate how diarrhea spleen deficiency occurrence in the mechanism of Glucose absorption; and in the treatment of by Buzhong Yiqi Decoction,which on the basis of each proportion of "Yiqi Shengyang" Drug compatibility, make sure the best dosage between astragalus-cimicifuga and bupleurum ". To excavate the scientific connotation of dosage based on the experiment, in order to guide the Clinical doctors for using.Methods:1 The effect of BZYQD each proportion of "Yiqi Shengyang" Drug compatibility on diarrhea spleen deficiency rats.Establish the spleen deficiency rat by the method of "food preferences+food control+exhaustive swimming" which were randomly divided into group "BZYQD" and its altered prescriptions-group "BZYQD with doubled cimicifuga and bupleurum" and group "BZYQD without cimicifuga and bupleixrum",to examine the body weight of rats, macro integration, thymus index and spleen index of the spleen deficiency rats by giving corresponding drug intervention.2 The effect of BZYQD each proportion of "Yiqi Shengyang" Drug compatibility on the repair of mucous membrane of small intestine of spleen deficiency rats.Establish the spleen deficiency rat by the method of "food preferences+food control+exhaustive swimming" which were randomly divided into group "BZYQD" and its altered prescriptions -group "BZYQD with doubled cimicifuga and bupleurum" and group "BZYQD without cimicifuga and bupleixrum", to observe the structure of the intestinal villi of the spleen deficiency rats through light microscope and the intestinal microvilli through transmission electron microscope,besides,measure the height and width of the small intestinal villi through the medical image analysis system by giving corresponding drug intervention.3 The effect of BZYQD each proportion of "Yiqi Shengyang" Drug compatibility on related gene GLUT2 belong the transport and absorption of glucose of diarrhea spleen deficiency rats.Establish the spleen deficiency rat by the method of "food preferences+food control+exhaustive swimming" which were randomly divided into group "BZYQD" and its altered prescriptions -group "BZYQD with doubled cimicifuga and bupleurum" and group "BZYQD without cimicifuga and bupleixrum"; using RT-PCR detection of related gene GLUT2, Na+-K+-ATPase and T1R2+T1R3, Cavl.3, PKC-β-Ⅱ subtype, belong the transport and absorption of glucose,come from the total RNA in the intestinal mucosa by giving corresponding drug intervention.Results:1 The body weight of rats, macro integration, thymus index and spleen index:The body weight of the rats was significantly decreased, and the macro integral significantly increased in model group (P<0.05). The thymus index and spleen index decreased significantly also in group model (P< 0.05);the body weight, thymus index and spleen index were significantly increased and the macro integral decreased in group "BZYQD" (P < 0.05),while the other two groups were not significantly changed on some indicators. (P>0.05)2 Pathological examination:the destruction of small intestine villi and Inflammatory cell infiltration,epithelial cell loss, destruction of cell crgan of the rats and the height and width of intestinal villi were significantly reduced (P<0.01) in the model group, after giving BZYQD,, the intestinal mucosal repair obviously,and the height and width of intestinal villi increased in group "BZYQD"(P< 0.01), while it has no significant change (P>0.05)in its altered prescriptions-group "BZYQD with doubled cimicifuga and bupleurum" and group "BZYQD" without cimicifuga and bupleixrum".3 The related genes belong the transport and absorption of glucose:the expression of gene GLUT2 and Na+-K+-ATP enzyme, PKC-beta Ⅱ subtype significantly decreased (P<0.05). beside of the gene TIR2, T1R3 and Cav1.3 (P<0.05)in model group.All the genes above-mentioned increased after treat by different group of BZYQD. Among that, group "BZYQD" were increased significantly (P<0.01) but its altered prescriptions -group "BZYQD with doubled cimicifuga and bupleurum" and group "BZYQD without cimicifuga and bupleixrum" did not (P>0.05).Conclusion:The change of general state of the rats with spleen diarrhea deficiency;the decline of the body weight, thymus index and spleen index decline; the destruction of the small intestine villi; the expression decreased on the related gene belong the transport and absorption of glucose, to authenticate the relation between the transport and absorption of glucose and the occurrence of diarrhea spleen deficiency,besides,its occurrence was relevant to the gene channel PKC beta Ⅱ-GLUT2-TIR2, T1R3/Cav1.3 in the intestinal mucous.The BZYQD can use for treating diarrhea spleen deficiency by protect the intestinal mucous enhance the expression of gene channel above-mentioned., and the BZYQD compatibility is the best especially.Further more,it confirmed the BZYQD "Yiqi Shengyang" Drug compatibility should for using the Yiqi drugs and Shengyang drugs, among that, large doses of drugs for Yiqi drugs and small doses of Shengyang drugs should give expression to the meaning of Yiqi Shengyang.
