Experimental Study On The Effects Of Supplemental Shengyang Yiwei Decoction In Treating Diarrhea-predominant Irritable Bowel Syndrome(The Type Of Liver Depression And Spleen Deficien)

Objective:The objective of this study was to establish a rat model of Diarrhea-predominant Irritable Bowel Syndrome,and choosing De Shute Pill as positive control group,observe the effect of Supplemental Shengyang Yiwei Decoction on rats gastrointestinal dynamics and visceral sensitivity,To explore the effect mechanism of Supplemental Shengyang Yiwei Decoction on Diarrhea-predominant Irritable Bowel Syndrome,as well as the role of prevention,treatment,and to provide more rigorous experimental basis for Supplemental Shengyang Yiwei Decoction in clinical widely be using and vigorously promoting.Methods:Fifty SPF Wistar male rats,weighted 180-200 g,Using random grouping method,rats were randomly divided into five groups including normal control group,model control group,Deshu Te Pill control group,higher-,lower-dose of Supplemental Shengyang Yiwei Decoction treatment groups,each 10 rats in each group.Choosing the way which is the constraint stress + senna + clip tail to make the model of IBS-D.Giving the normal control group with normal saline lavage(18 ml/kg)once per day.,don't put any stimulus through the whole process,giving the rest of the group rats all senna decoction lavage(18 ml/kg),and after an hour,using transparent tape constraints to limited them not to be optional in facial activity,but not effect their normal walking for an hour,At the same time,stimulating tail 15minutes;recording:the rats' hair luster,skin colour and lustre,emotional reactions and behavior state,increase or decrease of eating,drinking water,loose,weight increase or decrease in the process of stimulus;The forty day of mording,choosing two rats of each group of colon tissue,which was cleaned by 0.9% sodium chloride solution,to observe generally intestinal mucosa color,luster,to judge whether is any organization's congestion,including edema,exudation,erosion and other damage.To continue the stimulation,the third week of mording,respectively be Shute pill,higher-,lower-dose of Supplemental Shengyang Yiwei Decoction once per day for two weeks.Fasted rats for twenty four hour with 50%charcoal suspension,after thirty minutes to taking blood that each rat take four ml for biochemical experiment indexes such as: using immunohistochemical method to detect serum of rats at the end of the 5-HT,plasma VIP content,carbon distance and length of small intestine,and colon tissue to observe to have or not inflammatory lesions.The data was statisticed by SPSS18.0 for statistical processing.Results:1 General conditon Model rats compared with normal control group,there was reduce in diet at first week,individual mood irritability of rats is not stable,more difficult to grasp,to stimulate the resistance,compared with beginning of the experiment is less,but biting tail clip phenomenon continued to exist,the trapped after model rats keep higher alert,no phenomenon of the escaping at the beginning of the experiment;Model rats are loose at second week,rats are defecate pond,all big abdomen of fur are contaminated with feces,hair luster,activity significantly reduce,the shackles and clip tail stimulation obviously slow.2 The changes of weight Compared with normal group,the body weight of model group rat groups was all lighten after model(P<0.05).Two weeks after treatment,compared with model group,the body weight of treatment groups was increased,(P<0.05 or P<0.01).3 Behavior evaluation Compared with normal control group,the amount of sugar water consumption of model group rats was reduced gradually,more and more significant difference(P< 0.05).Each treatment group rats was increase gradually,which higher-dose of Supplemental Shengyang Yiwei Decoction had obvious difference.4 Pathological changes of colon There is no ciginificent changes of colon.Each group has no obvious ulcers,bleeding,erosion,and inflammatory cell infiltration.In a word,there is no significant differences among the groups.5 Compared the small intestine charcoal suspension pushing rate Compared with normal group,model group,Dicetel group and low dose group could increase charcoal suspension pushing rate,especially model group(P< 0.01).Compared with model group group,higher-dose of Supplemental Shengyang Yiwei Decoction had obvious difference(P< 0.05).6 The results of serum 5-HT,plasma VIP6.1 Serum 5-HT: compared with normal group,the content of serum 5-HT was significantly higher(P<0.01),compared with model group,the others groups of treatment were obviously had difference(P<0.05),but each group was not.6.2 Plasma VIP: compared with normal group,the content of VIP was significantly higher in model group(P<0.01),compared with model group,the others groups of treatment were obviously had difference(P<0.05 or P<0.01).Higher-dose group was obviously superior to other groups in reducing the expression of VIP(P<0.05).Conclusion: Supplemental Shengyang Yiwei Decoction could improve the general conditon of liver depression and spleen deficiency type of IBS-D rats.The mechanism is related to regulate the secretion and release of gastrointestinal hormone like 5-HT?VIP,to achieve the purpose of treatment IBS-D,and form apparente dose-effect in large dose.
