Object:The SD rat periodontitis model was constructed by combining the four methods of ligation method+high-sugar diet+Pg bacteria inoculation+prednisolone acetate injection.The effect of Yiqi Shengyang Guchi method on periodontitis was studied by using the Yiqi Shengyang Guchi Decoction to intervene SD rats.Methods:1.Establishment of experimental periodontitis model in SD rats:60 male and female SPF-grade SD rats of 8 weeks old were selected and divided randomly divide them into control group?20 rats?and model group?40 rats?according to their weight.After 2weeks of feeding adaptation,the model group was modeled with ligation of orthodontic wire in the neck of teeth,high sugar diet,inoculation of PG bacteria and prednisolone acetate injection on the right maxillary first molar of SD rats to establish chronic Periodontitis animal model.Four rats were sacrificed in each group at the fourth week of modeling,and periodontitis-related indicators,gingival index?GI?,sulcus bleeding index?SBI?,plaque index?PLI?,and periodontal pocket depth?PD?were measured.2.Study on the regulation of microenvironment immune response to experimental periodontitis in SD rats by the method of Yiqi Shengyang Guchi:50 SD rats were selected,16 of which were not modeled as the Control group;34 of which were modeled as the control group,were randomly divided into Model group?16 rats?,experimental group?YSGD group?18 rats.In the YSGD group,a solution of Yiqi Shengyang Guchi Decoction was orally administered at a concentration of 10.5 g/kg,administered once a day,and continuously administered 4 weeks.The Control group and the Model group were orally administered with an equal amount of normal saline,and the other feeding conditions were the same.Four SD rats in the Control group and Model group died at0W,1W,2W,and 4W in experiments,6 SD rats in the YSGD group died at 1W,2W,and 4W in experiments,and the first maxillary molar of the right side of SD rats was isolated Periodontal tissues were prepared for pathological sections.Immunohistochemical staining of cytokines IL-1?,TNF-?,PGE2,IL-6,and IL-17 was performed on periodontal tissue pathological sections,and quantitative image analysis was performed.The pathological changes of periodontal tissue were observed with HE staining.Results:1.The periodontitis model of SD rats was constructed by four methods of"ligation+high sugar diet+PG bacteria inoculation+prednisolone acetate injection".Model group had higher GI,SBI,PLI,PD than the Control group at 4W,and the difference was statistically significant?P<0.05?.2.HE staining of periodontal tissue in SD rats showed that the control group was normal periodontal tissue,no obvious abnormality was found;in the model group,the cells in lamina propria of mucosa were necrotic,with inflammatory cell infiltration.3.Regulating the HE staining of periodontal tissue SD experimental periodontitis in rats by the method of Yiqi Shengyang Guchi:At 0W,1W,2W,and 4W,the Control group showed normal periodontal tissues with no obvious abnormalities;0W,mild inflammatory cell infiltration was observed in the periodontal tissues of the Model group.At 1W,mild inflammatory cell infiltration was observed in the periodontal tissue of the Model group and the YSGD group.At 2W,moderate inflammatory cell infiltration was observed in the periodontal tissue of the Model group and YSGD group.At 4W,the periodontal tissue of the Model group was showed moderate inflammatory cell infiltration,and the YSGD group showed mild inflammatory cell infiltration.4.Study on the regulation of microenvironment immune response to experimental periodontitis in SD rats by the method of Yiqi Shengyang Guchi:Compared with the Control group,at 0W,1W,2W,and 4W,the expression of cytokines IL-1?,TNF-?,PGE2,IL-6 and IL-17 in the periodontal tissue microenvironment of the Model group increased?P<0.05?.Compared with the Model group,at 1W the expression of cytokines TNF-?,PGE2,and IL-6 in the periodontal tissue microenvironment decreased in the YSGD group,and the difference was statistically significant?P<0.05?;At 2W,the expression of cytokines TNF-?,PGE2,and IL-17 decreased in the environment,and the difference was statistically significant?P<0.05?;at 4W,the cytokines IL-1?,TNF-?,PGE2,IL-6 and IL-17 were all reduced,and the difference was statistically significant?P<0.05?.Conclusion:1.Four comprehensive methods of"ligation+high sugar diet+PG bacteria inoculation+prednisolone acetate injection"for the first grinding of the right upper jaw of SD rats can quickly and successfully construct the periodontitis animal model in a short time.2.Yiqi Shengyang Guchi Method can reduce the infiltration of inflammatory cells in periodontitis microenvironment.3.Yiqi Shengyang Guchi Method can effectively regulate the balance of immune response in the local microenvironment of periodontitis,and down regulate the expression of cytokines IL-1?,TNF-?,PGE2,IL-6,IL-17 in the microenvironment of periodontitis,and reduce periodontal tissue injury.
