Aims:The purpose of this paper isto analyze the IL-28B genetic variation in Yanbian population and its relevance with SNP and hepatitis C.Method:The allelic discrimination assay by Taqman real-time PCR was developed to determine genotypes of SNPs, rs12979860、rs8099917、rs12980275、 rs8105790, which were analyzed in190 hepatitis C patients and 104 healthy controls. The allele frequency between the patients with hepatitis C and healthy controls were compared to assess the effect of SNP, baseline virus load, liver function, and genotype on the treatment and prognosis of HCV.Result:The genotype frequencies of CC and CT of rs 12979860 in Chinese Korean with HCV infected were 90.82% and 9.18%,and 90.21% and 9.78% in Chinese with HCV infected;The genotype frequencies of TT and TG of rs8099917 in Chinese Korean with HCV infected were 91.83% and 98.16%,and 80.7% and 16.3% in Chinese with HCV infected; The genotype frequencies of AA and AG of rs12980275 in Chinese Korean with HCV infected were 89.8% and 10.2%,and 84.78% and 15.22% in Chinese with HCV infected;The genotype frequencies of TT and TC of rs8105790 in Chinese Korean with HCV infected were 91.83% and 8.16%,and 97.83% and 12.17% in Chinese with HCV infected. The four SNPs were in strong linkage disequilibrium with each other(r2>0.80). There was no significant difference between IL-28B SNP (rs12979860) genotype with HCV subtype, HCV-RNA and liver biochemical examination results.However, there were no significant difference between HCV infeceted group and health control group in the rate of genotype frequencies for four SNPs.Conclusion:The IL-28B SNPs gene phenotype of Chinese Korean and Chinese in Yanbian area are consistent with the phenotype in other parts of the region, and to no significant difference between the two group.There was no significant difference between IL-28B SNP (rs12979860) genotype with HCV subtype, HCV-RNA and liver biochemical examination results.That IL-28B SNPs gene detection were not enough to explain the susceptibility of Chinese Korean infected HCV.
