Objective:Through constructing Imiquimod-induced psoriasiform Inflammation of BALB/c mice back, detect the change of correction PASI score and histopathology of BALB/c mice, find the change of IL-18and IL-22level in BALB/c mice, evaluate the effect of Yinxieping pill to treat psoriasis, and explore the possible mechanisms.Methods:52healthful female BALB/c mice, weight ahout18to20grammes,age about8to12weeks, were randomly divided into six groups, including blank control group,model control group, Tripterygium glycosides group and Yinxieping pilll high, medium and low-dose group, each group of eight.Another mice are using for preliminary experiment.Only after the model successfully constructed,the experiment begins.Except blank control group with Vaseline coated outside, the other groups are used5%Imiquimod Cream coated one day at a time.At the same time by intragastric administration,the blank control group and model control group with0.4ml physiological saline,the Tripterygium glycosides group with0.4ml Tripterygium glycosides solution gastric lavage,the Yinxieping pilll high, medium and low-dose group with0.4ml high,medium,low dose of Yinxieping pilll solution gastric lavage one time a day for eight days.During the experiment, observe the changes of mice back skin lesions, the basic situation and record.24hours after the last administration, the mice back skin lesions were corrected PASI score.After the removal of the eyeball, blood0.8-1.0ml for detection of cytokines IL-18, IL-22,take back skin tissue for pathological section.Results:1.The mice back skin correction PASI scores of model control group, Tripterygium glycosides group and Yinxieping pill high, medium,low-dose group were higher than that of blank control group (P<0.05), and the difference was statistically significant.The mice back skin correction PASI scores of Tripterygium glycosides group and Yinxieping pill high, medium,low-dose group,were less than than of model control group (P<0.05), and the difference was statistically significant.2.IL-18and IL-22expression level in peripheral serum of model control group, Tripterygium glycosides group and Yinxieping pill high, medium,low-dose group mice,were higher than that of blank control group (P<0.05), and the difference was statistically significant. IL-18and IL-22expression level in peripheral serum of Tripterygium glycosides group and Yinxieping pill high, medium,were lower than that of model control group (P<0.05), and the difference was statistically significant.Conclusion:1.Yinxieping pill can reduce the PASI score and has certain therapeutic effect improvement to psoriatic skin lesions.2.IL-18and IL-22expression level in peripheral serum of imiquimod-induced psoriasis-like model BALB/c mice are higher than that of normal mice. There is an important role that elevated IL-18, IL-22expression level in peripheral serum for psoriasis pathogenesis.3.Yinxieping pill can reduce IL-18and IL-22expression level in peripheral serum of imiquimod-induced psoriasis-like model BALB/c mice, and influence the pathogenesis of psoriasis, and has certain intervention effect on the occurrence and development of psoriasis.
