Objective To observe the effects of sanxian capsule on HIF-1α,VEGF, AQP1expression in the Orthotopic xenograft model of renal cell carcinoma, and to explorethe role of the three in renal cell carcinoma.Methods The renal cell carcinoma786-0cells were cultured in vitro cellpassage, breeding.48nude mice were randomly divided into blank group, modelgroup, Sanxian capsule low, middle and high-dose group, interferon group.40nudemice were established of orthotopic renal carcinoma model,who were given per grambody weight0.0015g,0.003g,0.006g, dosing in nude mice for human administration30x, by converting the low, medium and high dose respectively. They were brood inthe SPF environment. The nude mice were observed diet,haircolor, mental status,activity and measured weight on every week. they were killed after4weeks offeeding. Kidney tissue biopsy was performed. Immuno-histo-chemistry SABCmethod was used to detect the expression of HIF-1α, VEGF, AQP1on the renalcell carcinoma tumor tissue,PBS instead of primary antibody as negative control. Thesamples were collected, formalin fixed, paraffin embedded. immunohistochemicalspecific operation according to the kit staining steps. HIF-1α staining: positive cellnuclei or cytoplasm was brown granular coloring. VEGF positive cells showed brownyellow granules stain. AQP1positive cells were yellow or brown granules stain.IPP6.0software screenshot were applicatd to detect the average optical density insection, after the application of statistical software were analysis variance methodfor statistical processing of date.Results Under light microscope observation showed: HIF-1α, VEGF have not expression on blank control group, AQP1has a litter expression. but three factorhave lots of expression in renal cell carcinoma. Statistical analysis of HIF-1α factordetection results are shown: model group compared with the blank group wasstatistically difference (p <0.05), indicating that the modeling success. San Xiancapsule group and interferon group are positive expression decreased obviously, therewas significant difference compared with model group (p<0.05), which confirmed:effective treatment.High dose group vs low, middle dose group had significantdifference (p<0.05). The Sanxian capsule high dose and interferon group hadsignificant differences (p<0.05). It confirm the high dose group curative effect isdistinct.VEGF detection showed: model group compared with the blank group wasstatistically difference (p <0.05), indicating that the modeling success.
