Objective:The aquaporins (AQPs) are a familyof small integral plasma membrane proteins that transport water and small molecules such as glycerol, urea, and even ions inspecial cases, across the plasma membrane. They are conserved in bacteria,plants, and animals. To date,13members of the AQP family have been identified in mammals. The nasal mucosa vascular endothelial growth factor is the strongest known vascular permeability factor, and its transparent effect is stronger than histamine1×106times.which has a strong role in the induction of vascular, hyperplasia formation of microvessels,at the same time the role of vascular endothelial cells regulating the permeation of the small blood vessels and capillaries that the plasma and some macromolecules (such as fibrinogen) was infiltrated into the extravascular matrix, resulting in tissue edema. The study found that the abnormal expression and regulation of loss of control of the either of AQP1and VEGF has a relationship with inflammatory, edema. few researehes had studied about them in allergic rhinitis.The meehanisms of allergic disorders were not clear. This study was designed to study the expressions of AQP1and VEGF in normal rats,rats of allergic rhinitis and application of dexamethasone in allergic thinitis rats, thus to evaluate the role of AQP1and VEGF in allergic rhinitis pathogenesis and the correlation of them.Methods:Thirty8-week-old healthy Wistar rats half male and half female, weight160g-200g were randomly divided into control group, allergic rhinitis group and dexamethasone therapy group after a week. There were ten rats in every group. The allergic rhinitis group and dexamethasone therapy group was induced by classic method with ovalbumin(OVA). Then,the therapy group was intraperitoneal injection of dexamethasone after the modeling for a week.The control group was deal with physiological saline instead by the same method. To observe the histomorphological characteristics of allergic rhinitis by Histochemical methods. Immumohistochemistry and real-time PCR methods were applied to detect the expression and the distribution of AQP1and VEGF in the level of protein and mRNA in nasal mucous membrane with AR.Result:According to the behavior assessment standards, the rats in model group and intervetion group were inspired typical symptoms of allergic rhinitis.The model of allergic rhinitis is successfully established. After the dexamethasone intervention,the rats in intervention group have occasional runny nose,sneezing and scratching nose phenomenon almost disappear. After the modeling, model group shown a significant increase of epithelial goblet cells, Infiltration of inflammatory cells, eosinophils, vessels and glands, the epithelial tissue oedema. After the treatment of dexamethasone, tissue edema, vascular dilation, gland hyperplasia and inflammatory cell infiltration were reduced. By symptomatology of rats the model group score higher than the control and intervention group. AQP1was expressed mainly on nasal mucosa coating the epithelial basement membrane, endothelial cell membrane and within the glandular cells, VEGF was expressed mainly on Small arterial and venous endothelial cells and glandular cells. AQP1and VEGF in the model group by immune histochemistry and real-time PCR assay were highly expressed in the intervention group was lower than the model group. The differences were statistically significant, and both have positive correlation.Conclusion:High expressions of AQP1and VEGF on the nasal mucosa of rats with AR indicate both of AQP1and VEGF might be pathological factors of AR that concered with gland’s excessive secretion and organization’s edema. Dexamethasone can down regulate the expression of them and reduce gland’s secretion and nasal obstruction by changing the expression of AQP1and VEGF. There was a positive correlation between AQP1and VEGF, but the exact mechanism is unclear...
