Objective:In this study, mice animal models were replicated to chloasma, Taohong siwu Decoction(THSWD) and other treatment to be observed before and after treatment in each group blood tyrosinase conntent and skin morphological pathology results, to discuss the effect of Taohongsiwu Decoction in the treatment of chloasma, hoping to provide further theoretical and experimental basis on clinical research about the prevention and treatment of chloasma.Methods:60white female mice of clean grade, weight about20g, were randomly divided into6groups (Blank group,Model group, THSWD prevention group, THSWD treatment group, Vc prevention group, VC treatment group),10in each group. Melasma animal models is made by UVB irradiation.Before the experiment, remove the hair from spine and buttock skin area of all mice by mild hair removal cream,clean the exposed skin on the back. Remove the hair every week,28days in total.All the treatment group gived orally drug therapy28days after28days UVB irradiated; the prevention group do not deal with anything in the early28days,on the after28days give them UVB irradiation and daily gavage of THSWD for immediate prevention. After the treatment detect the content of Tyrosinase in the blood, and observed the skin pathology morphology..Results:1. Based on the content of tyrosinase in the blood of mice in each group were analyzed, the blank group and model group in mice blood content of tyrosinase and skin pathology morphological differences, both have statistical significance (P<0.01). As the other4groups(THSWD prevention group, THSWD treatment group, Vc prevention group, VC treatment group) tyrosinase content decreased, compared with the model group, with statistical significance(P<0.01).This shows that the four groups of control has benign regulative action on pigmentation mice.2. According to the analysis of mice skin biopsy, the blank control group, epithelial superficial,without hyperplasia, acanthosis cell and basal cell is neat and orderly. The model group of mice model of hyperplasia of epithelial cells of the epidermis verrucous, papilloma significantly.showing more inflammatory cells, hair follicles, sebaceous glands and other skin appendages also appeared obvious hyperplasia. The THSWD prevention group mice skin compared with the blank group, slightly thickened skin, skin adnexal almost no hyperplasia, no inflammatory cells, epithelial hyperplasia of local slightly, the most obvious effect. The THSWD treatment group and the VC treatment group come second and third, the VC prevention group are not as good as the other three groups,, but the case has also been significantly improved with the model group.Conclusion: 1. The mechanism of preventing and treating the chloasma of THSWD may be reducing the content of tyrosinase,inhibiting the proliferation of melanocytes, thereby reducing the synthesis of melanin.2.Taohong siwu Decoction has significant effect on Melasma.3.
