Objective:In this study, mice animal models replicate chloasma, freckle Yurong Fang and other treatment to be observed before and after treatment in each group tyrosinase activity and skin blood morphological pathology results, and the mechanism of melasma combined were detected and analyzed to explore the freckle Yurong side effects on the treatment of melasma mechanism for further research, prevention and treatment of melasma provide theoretical and experimental evidence.Methods:48white female mice of clean grade, weight about20g, these mice were randomly divided into6groups (Model group, the Vc group, Chinese medicine group, Chinese medicine Plus mask group, Chinese medicine prevention group,blank group),8in each group. Melasma models is made by UVB irradiation.Before the experiment, remove the hair from spine and buttock skin area of all mice by mild hair removal cream,clean the exposed skin on the back. Remove the hair every week,4weeks in total. Apart from the blank control group, each group were exposed to the UVB wavelength of320nm for60minutes,once in a daily time. Because the skin of mice is very delicate, the UVB irradiation was divided in two phases,30minutes for each time and10minutes for cooling10minutes between. Keep the animals20cm away from the source of light. The UVB irradiation last for4weeks, observe the changes of the exposed skin of mice. After4weeks of UVB irradiation, treat each group with corresponding methods for4weeks.After the treatment detect the content of Tyrosinase in the blood, and observed the morphology of treated skin.Results:1. Analysis the content of Tyrosinase and skin pathology morphology of each group, there is significant difference in the normal group and melasma model group(P<0.01). The tyrosinase content of all treated group (Vc group, Chinese medicine group, Chinese medicine Plus mask group, Chinese medicine prevention group) were significant lower than model group, indicating that these four treatment method have effect on the skin pigment mice.2. Biopsy analysis shows that epithelial cell of blank control group is thinner, and non-proliferation, and the basal cells and prickle cells were arranged orderly. The epithelial cell of model control group show much proliferation and the increased number of epidermal cells, more inflammatory cells, and obvious hyperplasia of skin appendages such as hair follicles and sebaceous glands. The Chinese medicine plus mask group,compared with the blank control group, the epidermis is slightly thickened, the skin appendages are significantly reduced, only remaining small amount of inflammatory cells and hyperplasia fibrous tissue. Compared with the Chinese medicine group and Vc group, Chinese medicine plus mask group has the best treatment effect; preventive group ranked second and followed by Vc group, which is not as good as the first two treatment groups, but there has been significant improvement compared with the model group.Conclusion:1. The mechanisms of Qubanyurong may be reduce the content of tyrosine in skin and liver, antioxidant, improve SOD activity to inhibit the melanocyte proliferation, thereby reducing the the melanocyte synthesis.2. Qubanyurong treatment has significant effect on Melasma.3. Freckle Yurong side effect is obvious for the prevention of melasma...
