Objective: To investigate the relationship between Notch4 receptors and a ngiogenesis by blocking the Notch signaling pathway with DAPT,and then tra nsplanting the rabbit aut-ologous skin graft model with Taohong Siwu Decoctio n.Anastomotic wound surface,vasc-ular density detection and Notch4 receptor p rotein and mRNA content changes,revealing the effect of Taohong Siwu Decoct ion in the transplantation of rabbit skin graft model to promote the mechanism of angiogenesis.Methods: The rabbits were fed for 1 week and then numbered according to the order o-f weight,and finally grouped by random number table method.This study was divided into two groups,namely,?,?,respectively,I will be 60 experimental rabbits were divid-ed into four groups,namely,blockade group,model group,sham operation group and no-rmal control group,each group of15.DAPT subcutaneously was injected into the bloc-k after challenge,and th e other groups were given subcutaneous injection of DAPT solv-ent DMSO.Al l the rabbits were divided into four groups: the Taohong Siwu Decoction grou p,the model group,the sham operation group and the normal control group.A fter the model,the Taohong Siwu Decoction group was treated with Taohong Siwu Decocti-on,and the other groups were given the same amount of normal saline.All rabbits were randomly divided into two groups.The rabbits were s acrificed at 2d,5d and 8 d aft-er operation.The rabbits were sacrificed and the skin grafts of the grafted skin were cut The expression of Notch4 in the seru m was detected by ELISA,and the mRNA conten-t of Notch4 was detected b y RT-PCR.Immunohistochemistry was used to detect the ex-pression of Notch4 receptor in the immunohistochemistry group.Optical density.Results: The vascular density of rabbit skin grafts in group?was significa ntly lower tha-n that in model group(P<0.05),and the difference was statistica lly significant.The prot-ein and mRNA content of Notch4 in transplanted skin was lower than that in model gr-oup Model group(P<0.05),the difference wasstatistically significant.(P<0.05),the diffe-rence was statistically significant.The protein and mRNA content of Notch4 in the tran-splanted skin of Taohong Si wu Decoction was higher than that in the model group(P<0.05),and the diffe rence was statistically significant Group(P<0.05),the difference was statisticall y significant.Conclusion: 1.DAPT blocking Notch signaling pathway can inhibit the e xpression of N-otch4 receptor and affect angiogenesis.2.Taohong Siwu Decocti on can promote the angi-ogenesis by regulating the expression of Notch4 recep tors in Notch signaling pathway.
