Objective: To investigate the expression of Jagged1 ligand protein,the expression of Jagged1 ligand gene and the vascular density in the anastomotic area of the Notch signaling pathway in the serum by using the rabbit skin graft model as the research object.To explore the relationship between Taohong Siwu Decoction,Jagged1 ligand and angiogenesis.Methods: 120 experimental rabbits were fed for 1 week then divided into two parts,and the male and female were 50% in each part.Each part should numbering according to body weight from light to heavy order,Finally,have been divided into five groups for using the random number table.The first part: Blocking group,model group,sham operation group,normal control group.each group of 15.After operated separately,the rabbits were sacrificed and taken blood at the time of 2,5 and 8 days.The rats in the model group and the inhibitor group were cut the transplantation skin graft,while sham operation group and normal group The control group in the corresponding bit to be taken the same thickness of the skin.The protein content of Notch/Jagged1 in serum was detected by ELISA.The expression of Jagged1 gene was detected by RT-PCR,and the corresponding vascular density was detected by immunohistochemistry.The second part of the group was Taohong Siwu Decoction group,model group,sham operation group and normal control group,each group of 15.The rats in each group were sacrificed at the time of 2,5 and 8 days after operation.Five rabbits were randomly selected from each group.The rats in the model group and the blocking group were treated with transplantation skin graft,sham operation group and normal group The control group in the corresponding bit to be taken the same thickness of the skin.The protein content of Notch/Jagged1 was detected by ELISA,and the expression of Jagged1 gene was analyzed by RT-PCR.Results:(1)The first part of the experimental data showed that the density of neovascularization in the grafts was significantly lower than that in the other three groups in 5?8days(P <0.05).The difference was statistically significant.The expression of Jagged1 ligand in serum and the expression of gene expression in graft area were lower than those in the other three groups(P <0.05).The difference was statistically significant.(2)The second part of the experiment by experimental data analysis showed that the second day after surgery,Comparison between the numerical groups of Jagged1 ligand in the serum concentration and grafted skin anastomosis gene expression values.The values of Taohong Siwu Decoction group were higher than those of model group,sham operation group and normal control group(P <0.05).Comparison within the group shows that the results of 2d's of Jagged1 concentration in the serum of the Taohong Siwu Decoction group and the model group were significantly higher than those in the control group Compared with 5d's,8d's,P <0.05,the difference was statistically significant(P <0.05);the Comparison of 5d's and 8d's was not statistically significant(P> 0.05).Conclusion:(1)In the rabbit skin graft model,the expression of Jagged1 ligand can effectively increase the vascular density of the epidermis anastomosis.(2)In the rabbit skin graft model,the Taohong Siwu Decoction can effectively increase the expression of Jagged1 in the blood vessels,and when the vascular density reaches certain,the Taohong Siwu Decoction has no effect.
