| Objective:Through rabbit skin graft model as the research object, detection of effect of Taohongsiwu Decoction on angiogenesis promoting factor of serum Ang concentration and gene expression, to investigate the effect of Taohongsiwu Decoction on promoting angiogenesis skin graft.Methods:75rabbits, adaptive feeding for1weeks after weighing, then according to the weight from light to heavy sequence number, and finally the use of randomly divided into5groups, namely, Taohong Siwu Decoction low dose group, middle dose group and Taohongsiwu Decoction, Taohong Siwu Decoction high dose group, model group and sham operation group,15rats in each group. After modeling were orally, and postoperative2days,5dsys,8dsys three time, from each group were randomly selected5rabbits, heart blood after be executed, And cut the skin grafts anastomosed area skin tissue. By detecting the concentration of ELISA in serum samples detection method Ang, mRNA to detect Ang content in RT-PCR detection method.Results:(1) after the assay data analysis, Taohongsiwu Decoction group of skin grafts of microvessel density was significantly better than the model group (P<0.05), the difference was statistically significant. Ang expression in experimental animals show Taohongsiwu Decoction of three groups of expression is higher than that of the model group and operation group (P<0.05), the difference was statistically significant.(2) After the assay data analysis, Taohongsiwu Decoction group, middle dose group, high skin microvessel density and the expression of Ang is obviously better than the low dose group (P<0.05), the difference was statistically significant.(3) Taohongsiwutang high dose group and middle dose group skin expression of microvessel density and Ang, the difference was not statistically significant.Conclusion:Taohongsiwu Decoction can promote angiogenesis upregulation of tissue factor expression of Ang, as the classic prescriptions of promoting blood circulation and removing blood stasis, formula simplification, efficacy significantly, worth clinical promotion. |
