ObjectiveTo Explore the effect of Chaishao Chengqi Decoction on microcirculation ofsevere acute pancreatitis patients in observing the Concentration variation ofendothelin-1(ET-1), NO, GMP-140, TXB2and vWF:Ag before and after treatment.Methods100standard SAP patients were randomly divided into group A (50) and group B(50), two groups of patients in terms of gender, age, condition, duration have nostatistical significance (P>0.05), another30healthy volunteers were selected as acontrol group. A group with conventional therapy, B group with conventional therapy+Chaishao Chengqi decoction.By radioimmunoassay (RIA) for determination ofserum endothelin (ET-1). Serum nitric oxide (NO) content was measured by nitratereductase method. Serum Platelet alpha granule membrane protein (GMP-140) andthromboxane B2(TXB2) were test by ELISA method. the concentration of Serumvon Willebrand factor related antigen (Vwf:Ag) was test by monoclonal enzyme-linked immunosorbent assay.ResultsThe concentrations of GMP-140, TXB2,vWF:Ag, ET-1of SAP patients [groupA:56.12±16.73,97.36±35.14,169.59±40.48,60.37±11.23;group B:54.63±15.14,93.15±36.74,173.49±45.31,58.24±10.25] were significantly higher than those in thecontrol group[41.66±10.2,69.43±26.82,104.27±23.61,12.52±9.12](P<0.01);andthe Serum NO concentrations of SAP patients[group A:37.33±5.2,group B: 35.51±4.5] were significantly lower than the control group [59.62±3.72](P<0.05);After conventional treatment, the concentrations of GMP-140, TXB2vWF:Ag, ET-1of group A[51.81±13.47,93.58±34.16,42.12±5.38,160.52±34.18,55.36±9.15]has no significant difference compared with those before treatment[56.12±16.73,97.36±35.14,37.33±5.22,169.59±40.48,60.37±11.23](P>0.05); but theconcentrations of GMP-140, TXB2vWF:Ag, ET-1of group B[41.16±11.28,50.52±4.81,119.51±33.66,23.58±11.14] which with conventional therapy+Chaishao Chengqi decoction were significantly lower compared with those beforetreatment[54.63±15.14,93.15±36.74,173.49±45.31,58.24±10.25](P<0.05); and theconcentrations of Serum NO of group B[71.83±31.16] which with conventionaltherapy+Chaishao Chengqi decoction were significantly higher compared with thosebefore treatment[35.51±4.51](P<0.05)ConclusionsSerum NO,ET-1, GMP-140, TXB2and vWF:Ag participated in the developmentprocess of AP microcirculation. Chaishao chengqi decoction can reduce ET-1、GMP-140、TXB2、vWF:Ag and adjusting the balance of the NO/ET-1by this way,it can reduce microvascular spasm, Increase the perfusion of microcirculation, andInhibiting the formation of micro thrombosis,then improve the microcirculation ofSAP patients. improve the prognosis and the cure rate of SAP.
