Research On Therapeutic Formula Of Andrographis-isatidis Oral Liquid In The Early Of Warm Disease

Objective: There are many traditional Chinese medicine in treatment of early warm disease, which were contained in the Chinese Veterinary Pharmacopoeia like "Qingjie oral liquid", "Qingwen Baidu San" and "Yinqiao Powder" and "Shuanghuanglian oral liquid". These prescriptions were restricted in the animal husbandry because of high production cost. In order to select the cheap and effective prescriptions, Radix Isatidis, Folium isatidis and Andrographis paniculata were combined to a new prescription. In this experiment, the preparation process, pharmacodynamics, quality control and toxicology were studied to select the qualitive and cheap prescription for early warm disease.Method:(1) Based on the qualitity of R, S-Goitrin, Andrographolide, dehydroandrographolide, the better extriction method and prescription were selected(2) Three Andrographis-Isatidis oral liquid samples wre prepared to prevent and treat warm disease model of mice in order to determine the best prescription of Andrographis-Isatidis oral liquid.(3) TLC identification and quality determination of Radix Isatidis, Folium isatidis and Andrographis paniculata were studied to control the quality of Andrographis-Isatidis oral liquid.(4) Acute toxicity test and Long term toxicity test of mice were studied to evaluate the toxicity of Andrographis-Isatidis oral liquid.Results:(1) In the preparation experiment, extact of Radix isatidis and Folium Isatidis were obtained by Water extraction and alcohol precipitation. Andrographis paniculata was extracted by warm 75% Ethanol. The dedermined index were highest when Radix Isatidis, Folium isatidis and Andrographis were mixed in the proportion of 2:1:1.(2) In the pharmacodynamics experiment, it had the best antipyretic effect when extract of dehydroandrographolide, Radix Isatidis, Folium isatidis and Andrographis were mixed in the proportion of 2:1:1.(3) TLC of Radix Isatidis, Folium isatidis and Andrographis paniculata, and quality determination of Folium Isatidis and Andrographis paniculata were studied.Results showed that the methods don’t have negative diturb.It could be used as the quality control method of Andrographis-Isatidis oral liquid.(4) In the acute toxicity test, the mice didn’t die in senven days at the dose of 40000mg·kg-1, Anatomical characteristics were not obvious. In the long term toxicity test, The added weight, Routine blood test and Blood biochemistry Indexs of the rats were not obvious.Besides, Necropsy and histopathological examination wre not obvious, it showed that Andrographis-Isatidis have no obvious toxicity.Conclusion: The preparation of Andrographis-Isatidis oral liquid was simple and quick. The antipyretic effect is significantly.The determination were feasible and stable, it could control the quality of Andrographis-Isatidis oral liquid effectively. This precripiton had not obvious toxicity, so it could be developed as a new drug.
