Fufangqiancaoguanzhuye was effectively constituent preparation and formula of Rubia Cordifolia L., Salvia miltiorrhiza Bge. and Sophora Flavescens Ait. by using the methods of the modernization of traditional Chinese medicine in order to achieve the best effect of cow mastitis treatment. Fufangqiancaoguanzhuye had antibacterial antiphlogistic, promoting blood flow etc. The extraction orthogonal experiment of Rubia Cordifolia L., the pharmacodynamics and toxicology of fufangqiancaoguanzhuye were studied.1. The preparation of alcohol extract from RubimaillinThe content of Rubimaillin was used as target to conduct heat reflux extraction process in order to search for the best method by means of orthogonal experiment whose influencing factors are extraction time, ethanol concentration, solvent consumption and extraction frequency. The results showed that the optimal conditions were using 8 times of 70 % alcohol and heating reflux extraction thrice last for 2.5 h in the heat reflux extraction process.2. Study on PharmacodynamicsRubia Cordifolia L., Salvia miltiorrhiza Bge., Sophora Flavescens Ait and fufangqiancaoguanzhuye were conducted respectively to observe their the antibacterial effect in vitro. The result showed that the bacteriostatic effect of fufangqiancaoguanzhuye is better than the single traditional Chinese medicine. Three minimum inhibitory concentrations showed that the bacteriostatic effect of fufangqiancaoguanzhuye is better than the other two compound control groups. The results hinted that fufangqiancaoguanzhuye had good antibacterial effect.3. Study on ToxicologyLD50 of the mice was 6.59 g/kg in acute toxicity tests. And this dose was suggested the fufangqiancaoguanzhuye should be actual asepsis on the basis of herbal toxicity classification standards. In the chronic toxicity tests, rats of Wistar were lavaged the fufangqiancaoguanzhuye of 1/5 LD50, 1/20 LD50, 1/80 LD50 and zero dose to observe the clinical manifestations. Physiological, biochemical blood test, visceral index, pathohistology, etc were determinated after experiments. The results showed that there were significant differences as compared with the control group. 21 days dosage increase by degrees test design was done in the accumulation toxicity test of mouse, and no death until the end of experiment. It showed that fufangqiancaoguanzhuye was non-accumulation toxicity. 30 mice were given half lethal dose of death once. 13 mice died in all. It suggested that fufangqiancaoguanzhuye did not produce resistance. The average values of rabbit eye irritation were between 0 and 4 in the eye irritation test. It showed that the fufangqiancaoguanzhuye was no ritation.
