| Necrotic enteritis caused by C.perfringens mainly infects broilers aged 2 to 6weeks,with a high mortality rate,causing substantial economic losses to the breeding industry.Antibiotics usually used to prevent NE caused by C.perfringens.The unreasonable use of antibiotics has led to the emergence of bacterial resistance and the problem of food drug residues.Therefore,developing new antibacterial drugs against C.perfringens infection is urgently needed.TFP is essential for host-pathogen interaction and colonization.Screening inhibitors targeting C.perfringens TFP system has become a crucial anti-infection strategy.Compared with antibiotics,traditional Chinese medicine(TCM)has the advantages of comprehensive source,low price,diverse activity,and no drug residue,which provides valuable resources for developing new drugs.Based on previous studies,the natural compound galangin with good activity was obtained by screening inhibitors targeting C.perfringens TFP system.Our research group established targeting TFP to screen natural compounds and obtained galangin with good training.AOHOL was developed through prescription optimization,preparation process,quality standard research and stability investigation which is convenient for group administration in the chicken industry.Investigated the preventive effect of AOHOL on NE caused by C.perfringens infection in broilers was through clinical trials.The main results are as follows :(1)Firstly,the alcohol extraction process of galangin in Alpinia officinarum Hance optimized by orthogonal test.The excipients screened to determine the prescription of AOHOL: 30 g of Alpinia officinarum Hance was crushed,five times of 80% absolute ethanol was added,refluxed twice,2 h/time,the filtrate was combined.The relative density was above 1.04,15 m L of N-N dimethylacetamide,5m L of polysorbate 80,1 m L of vitamin E,PEG400 was added to 100 m L,mixed,sterilized,filled and sealed.High performance liquid chromatography(HPLC)was used to determine the content of galangin in AOHOL and carried out the methodological verification.The method has high sensitivity,strong specificity,good reproducibility,and good linear relationship.The influencing factor test,accelerated test,and a long-term test showed that AOHOL was suitable for storage at room temperature and cool environment,and the validity period was 12 months.(2)AOHOL(calculated by galangin)could inhibit the sliding of C.perfringen ATCC13124 in the concentration range of 5~80 μg/m L.5~80 μg/m L AOHOL(calculated by galangin)did not significantly inhibit the growth of C.perfringens ATCC13124.The MIC of AOHOL(calculated by galangin)to C.perfringens ATCC13124 was more than 160 μg/m L.5~80 μg/m L AOHOL(calculated by galangin)can effectively inhibited the biofilm formation and adhesion of C.perfringens ATCC13124.(3)AOHOL can restore the mental state of infected broilers,alleviate the pathological intestinal damage of infected broilers,and improve the production performance of broilers,suggesting that AOHOL has an excellent preventive effect on NE in broilers.The AOHOL developed stable content and a sound effect,and revealed its primary mechanism of inhibiting TFP function.It has an excellent preventive impact on NE in broilers and provides options for rhe clinical prevention of NE. |
