Objective:The anti-inflammatory effects of Qi Wei Li Fei Tang on Sprague-Dawley rat bronchial asthma model in vivo were evaluated from aspects of ethology, lung function, pathology of lung tissue, inflammatory cells and expression of NF-κB/p65and IκB in lung tissue. The mechanism of Qi Wei Li Fei Tang attenuating inflammation of bronchial asthma was further explained.Method:70SD rats were randomly divided into seven groups:control group, model group, blocker group, dexamethasone group, low-dosage group, middle-dosage group, high-dosage group. Ethology symptom was observed during OVA stimulated phase. Total and differential leukocytes counting in BALF were determined under the optical microscope. HE staining pathological section of upper lobe of right lung tissue was made to observe the pathological change and average alveolar area. ELISA was applied to quantify IgE in peripheral blood. The expression of NF-KB/p65and IκBαProtein in middle lobe of right lung tissue were detected by Western Blot. Real-Time PCR was used to test the express level of NF-KB/p65and IκBα in lower lobe of right lung tissue.Result:1. BALF:Preliminary observation the number of them from high to low was the model control group,the Chinese medicine low-dosage group,the Chinese medicine middle-dosage group,the Chinese medicine high-dosage group,the western medicine group,the blocker control group,the blank control group.Count results showed that:Compared with the blank control group,the result of the model control group’s BALF smear EOS count was significant elevated (P<0.01);the blocker control group’s was a little higher(P>0.05,no statistically significant);the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were higher(P<0.01) and showed the concentration-response relationship;and the western medicine group’s was higher too(P<0.01).Compared with the model control group,the blocker control group’s was lower (P<0.01);the Chinese medicine low-dosage,middle-dosage and high-dosage groups’were lower too,but the low-dosage group’s P>0.05(no statistically significant),when the middle-dosage and high-dosage groups’ P<0.01;and the western medicine group’s was lower(P <0.01).Compared with the blocker control group,the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were higher(P<0.01);and the western medicine group’s was higher too,but P>0.05(no statistically significant).Compared with the western medicine group,the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were higher,but the high-dosage group’s P>0.05(no statistically significant),and the low-dosage,middle-dosage groups’ P<0.01.the results of BALF smear EOS count from high to low among every group was the model control group,the Chinese medicine low-dosage,middle-dosage,high-dosage groups,the western medicine group,the blocker control group,the blank control group 2. HE stain:The blank control group had not showed any change and inflammatory cellular infiltration,when he blocker control group had a little,and the other groups all had inflammatory cellular infiltration in the lung tissue,bronchial epithelial cells irregular arrangement and desquamation,thic-kened alveoli septum and bronchiolar spasm,but the pathological damage severity were different.Results of the score by using tissue injury and inflammation pathological scoring criteria showed that:On the item of the inflammatory cellular infiltration,the score from high to low was the model control group=the Chinese medicine low-dosage group> the Chinese medicine middle-dosage group=the Chinese medicine high-dosage group> the western medicine group> the blocker control group=the blank control group.On the item of the bronchial epithelial cells desquamation, the score from high to low was the model control group> the Chinese medicine low-dosage group=the Chinese medicine middle-dosage group> the Chinese medicine high-dosage group=the western medicine group> the blocker control group=the blank control group.On the item of the thickened alveoli septum, the score from high to low was the model control group=the Chinese medicine low-dosage group> the Chinese medicine middle-dosage group> the Chinese medicine high-dosage group=the western medicine group> the blocker control group=the blank control group.On the item of the bronchiolar spasm, the score from high to low was the model control group=the Chinese medicine low-dosage group> the Chinese medicine middle-dosage group=the Chinese medicine high-dosage group> the western medicine group> the blocker control group=the blank control group.And the total points from high to low was the model control group=the Chinese medicine low-dosage group> the Chinese medicine middle-dosage group> the Chinese medicine high-dosage group> the western medicine group> the blocker control group> the blank control group.all above P<0.01.3. IgE in serum:Compared with the blank control group,every group’s serum IgE content increased (P<0.01),and the Chinese medicine low-dosage,middle-dosage and high-dosage groups showed the concentration-response relationship.Compared with the model control group,the blocker control group’s was lower (P<0.01);the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were lower too,but the low-dosage group’s P>0.05(no statistically significant),when the middle-dosage and high-dosage groups’ P<0.05;and the western medicine group’s was lower(P<0.01).Compared with the blocker control group,the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were higher(the low-dosage group’s P<0.05,while the middle-dosage and high-dosage groups’ P<0.01);and the western medicine group’s was higher too,but P>0.05(no statistically significant).Compared with the western medicine group,the Chinese medicine low-dosage,middle-dosage and high-dosage groups’ were higher,but the high-dosage group’s P>0.05(no statistically significant),and the low-dosage group’s P<0.01,the middle-dosage group’s P<0.05.4. RealTime-PCR:The expression of p65mRNA between each groups have no statistically significant. The model group is the highest one. The expression of IκBα mRNA between each groups have no statistically significant.5. Western Blot.The analysis of expression of p65nuclear protein showed, compared with control groups the relative expression amount of the other groups is increased (P<0.05); compared with model groups the relative expression amount of the other groups is decreased (P <0.05),dexamethasone group, blocker group and high-dosage group decreased notable (P<0.01); compared with blocker groups the relative expression amount of high-dosage group is increased((P<0.05), middle-dosage and low-dosage group increased notable (P<0.01); high-dosage group, middle-dosage group and loe-dosage group showed concentration-response relationship. The analysis of expression of IκBα nuclear protein showed compared with control groups the relative expression amount of the other groups decreased(P<0.05); compared with model groups the relative expression amount of the other groups is increased (P<0.05)Conclusion:1.Qi Wei Li Fei Tang can relief asthma effectively, hish-dosage is best.2.Under this experiment, the anti-inflammatory effect of dexamethasone is better than Qi Wei Li Fei Tang, however no side effect was observed in Qi Wei Li Fei Tang group.3. Qi Wei Li Fei Tang can up-regulate p65in nuclear and down-regulate IκBα in cytoplasm in translation level.4. Qi Wei Li Fei Tang’s anti-inflammatroy effect is associated with its suppressive effect on NF-κB signalling pathway.
