TCM have more obvious advantages and characteristics in terms of improving the quality of life of patients."Qi Wei Li fei Tang" roots from the principles of "Wen Run Xin Jin Pei Ben", which base on the research platform of "The Key Institute of state Administration of Traditional Chinese Medicine", have a good basic research evidence through continuous curing and standardization.Oxidative stress is when the body is in a pathological state, free radicals produce too much or too little, or clear radical enzymes, or both coexist, homeostasis is destroyed cause irreversible oxidative damage, eventually leading to the destruction of cell structure and function. Studies have shown that oxidative stress is closely related to the development and progression of asthma. Oxygen free radicals and antioxidant enzyme involved in bronchial asthma mainly NO、·OH、O2-·、SOD、MDA and GSH-PX.Objective:By observing the "Qi Wei Li fei Tang" intervention asthmatic rats, oxidative stress indicators in serum、lung homogenates and plasma (O2-·、NO、OH、SOD、MDA and GSH-PX) changes in circumstances, reveal antioxidant mechanisms "Qi Wei Li fei tang" treatment of bronchial asthma.Methods:SPF male SD rats were divided into:normal control group, model control group, low、middle and high dose group and western medicine group, adaptive feeding after7d, on dl, d8to five points (abdominal cavity, Bilateral the toes, Bilateral inguinal subcutaneous) injection of10%OVA/Al (OH)3+0.0023‰pertussis toxoid suspension of lml sensitization (each at0.2ml); From d15to start with1%OVA saline inhalation excited every day once,30min/times of excited7d. The normal control group received saline injection and atomization. At d9(after the completion of the last five-pointinjection) started gavage, animals were gavaged1/d, of14d. the low, medium and high dose groups, respectively, per day to be gavaged of0.182g/ml,0.363g/ml,0.727g/ml of aqueous extract (low-dose group was fed daily medium dose groups dose1/2, the high-dose group was fed daily medium dose group dose2-fold); medicine daily to be dexamethasone group was50μg/ml,3ml. Blank control group, model control group received saline daily.After treatment, Indicators with pulmonary function testing, bronchial lung cells lavage inflammatory cell counts, blood IgE count to evaluate animal model; using electronic paramagnetic technology and other methods detect rats serurm、lung homogenates and plasma oxygen free radicals and antioxidant enzyme involved in bronchial asthma mainly NO、·OH、 O2-·、SOD、MDA and GSH-PX.Results:1Model evaluationThe general performance, Model control group rats showed widespread wheezing, shortness of breath, lips white, cough, appearance of visible by fur lackluster, weight loss and other symptoms. Part of the Normal control group of rats shortness of breath.Pulmonary function test results, Model control group AHR (I) higher than the Normal control group (P<0.05); while the inspiratory capacity (IC), tissue damping (G), the organization hysteresis (H) etc. higher than the control group, but the difference was not statistically significant (P>0.05);Pulmonary histopathology results that Normal control group bronchial wall smooth, wrinkle clear rules; alveolar complete and regular; Model control group had bronchial basement membrane thickening and morphological changes; beside the obvious visible blood vessels and bronchial inflammatory cell infiltration, bronchial epithelial cell shedding; alveolar collapse, fusion;BALF results, EOS and neutrophils of Model group was higher than the Normal control group (P<0.05);The results for radical scavenging:Model group, serum free radical scavenging ability was lower than the Normal control group (p<0.05).2Serum oxygen radical scavenging resultsThe serum levels of NO scavenging difference was not statistically significant compared (P>0.05). Among them, the control group, Chinese high-dose group, low dose group cleared NO capacity than western medicine group (p<0.05); the serum for·OH and O2-· Clear the ability to compare the difference was not statistically significant (P>0.05).3Tissue oxygen radical scavenging resultsThe lung tissue showed no ability to remove NO difference (P>0.05). Among them, the Chinese high-dose group cleared NO capacity than western medicine group (P<0.05). The ability of lung tissue homogenates Clear·OH and O2-·1s. was no difference (P>0.05).4The results of plasma on SOD, MDA, GSH-PX Activity after the medication4.1Have more SOD activity in plasma levels of each group differences (p<0.05). Blank control group and Chinese high-dose group compared to the differences in plasma SOD activity (p>0.05), the control group SOD higher than the other groups (p<0.05); Model group plasma SOD activity was significantly lower than the other groups (p<0.05); medicine plasma SOD in the high dose group was significantly higher than western medicine group (p <0.05), western medicine group compared with the low dose group of SOD was no significant difference (p>0.05);4.2Plasma levels of MDA vitality compare the results statistically significant difference between groups (p<0.05). The blank control group comparison of plasma MDA medicine vitality levels in each group had no significant difference (p>0.05). Blank control group, the plasma levels of MDA vitality lower than the model group, western medicine group (p<0.05); The plasma MDA levels showed no difference with model group and western medicine group (p>0.05). Chinese high-dose group, middle dose group medicine plasma MDA levels lower than western medicine group activity (p<0.05). Low-dose group and western medicine group was no difference in plasma levels of MDA activity (p>0.05).4.3There was no difference in plasma levels of GSH-PX activity results in each group (p>0.05). Blank control group GSH-PX activity in plasma levels higher than the model group(p<0.05). Western medicine group in plasma GSH-PX activity higher than Chinese medicine group, but the difference was not statistically significant (p>0.05).Conclusion:1By ovalbumin, aluminum hydroxide and pertussis toxin five-point suspension ovalbumin sensitization and atomization stimulate methods can be successfully established bronchial asthma animal model.2In the occurrence and development process of bronchial asthma,the body’s oxygen radical scavenging capacity decreased,and oxidative stress participate in the development and progression of asthma.3"Qi Wei Li Fei Tang" can be a good clearance of serum and lung tissue homogenates of NO radicals, but has no effect on·OH and O2-·scavenging capacity; And can increase serum SOD levels, lower levels of MDA, and plasma GSH-PX on the role was not significant."Qi Wei Li Fei Tang" may influence the body’s oxygen free radicals and antioxidant levels of antioxidant enzymes play a therapeutic purposes asthma.
