| Objective:To study the correlation between HIF-1αand oxidative stress state and the onset of asthma by detecting the levels of serum HIF-1αand MDA,the activities and content of SOD,and GSH-Px activities in asthmatic patients,explore the role of HIF-1αand oxidative stress levels in bronchial asthma.Methods:A total of 45 patients with bronchial asthma who were in the outpatient clinics and hospitals of our hospital from August 2020 to February 2021 meeting the inclusion criteria were collected.Among them,30 patients(18 females and 12 males)were in the asthma attack group,and 15 patients(10 females,5 males)were in the asthma alleviation group.15 healthy adults(7 females and 8 males)for physical examination in our hospital served as a normal control group during the same period.Recording the corresponding blood eosinophil count,p O2,and lung function indicators(including FEV1%pred,FEV1/FVC value)in asthma patients when they were admitted to the hospital.The peripheral blood of all subjects was collected,and the serum was collected after centrifugation at 3500 r/min and stored in a refrigerator of-70°C.Enzyme-linked immunoassay(ELISA)was used to detect HIF-1αlevels and SOD content in serum of all subjects.the activities of SOD and GSH-Px in serum were respectively detected by MDA,SOD and GSH-Px kits in serum of all subjects to evaluate the level of oxidative stress and antioxidant function in asthmatic patients.Results:1.serum HIF-1α and MDA in acute attack of asthma group were evidently higher than those in the remission group(p<0.05)and healthy adults group(p<0.05),and the HIF-1αand MDA level in the group of asthmatic patients in paracmasia were evidently higher than those in the healthy adults as control group(p<0.05).2.Comparing with the group of asthmatic patients in paracmasia(p>0.05)and healthy adults(p>0.05),there was no significant difference in serum SOD and GSH-Px enzyme activities in acute attack of asthma group were higher,while content of SOD were lower(p<0.05);There was no significant difference in serum SOD and GSH-Px enzyme activities between the healthy adults group and asthma remission group(p>0.05),while content of SOD in remission group were lower than healthy adults group(p<0.05).3.FEV1%pred,FEV1/FVC had a positive correlation with the level of serum HIF-1αand MDA in asthmatic patients(p<0.05).4.Serum HIF-1αlevels in patients with asthma is positively correlated with MDA(p<0.05),partial pressure of oxygen is negatively correlated with HIF-1αand MDA levels(p<0.05),and the number of peripheral blood eosinophils had a positive correlation with HIF-1α,MDA level(p<0.05).Conclusion:1.Serum of HIF-1α and oxidative stress levels in asthmatic patients are significantly increased,and the levels in the attack phase are higher than those in the remission phase.HIF-1αand oxidative stress promote the development of asthma.2.HIF-1α and MDA levels has a positive correlation in asthmatic patients,while HIF-1αand SOD levels are negatively correlated,suggesting that there is an oxidative imbalance in asthma patients.HIF-1αand oxidative stress have a synergistic effect on the aggravation of asthma,and the two together promote the sustained attack of asthma.3.The lower the partial pressure of oxygen in the blood of patients with asthma,the higher the levels of HIF-1αand MDA,the worse the lung function,and the more severe the disease.4.The levels of HIF-1α and oxidative stress in asthmatic patients are positively correlated with the number of eosinophils,indicating that hypoxia and oxidative stress can promote airway inflammation in asthma by increasing the number of eosinophils. |
