On Acute Soft Tissue Contusion Of IL-1β And Histological Effects Of Four Cyperus Scattered

To use the method of taking orally Sizhixiangfu-san of ProfessorZheng Huaixian who was the late well-known export in our hospital, to explore theeffects of Sizhixiangfu-san on IL-1β and histological changes in rats with acute softtissue contusion and the possible mechanism, and to provide the theoretical basis for itsclinical use and lay the foundation for systematic study of Prof. Zheng Huaixian’sShangke Recipe system.Methods:72SD rats who were combated to set up the model of acute soft tissuecontusion. They were randomly divided into three groups (n=24), named asexperimental group (Sizhixiangfu-san), drug control group (Qili-san) and blank controlgroup, and then each group was randomly divided into four time point groups (n=6),named as the first day group, the third day group, the sixth day group, the tenth daygroup. At modeling an hour later, the rats were given medicine or normal saline bylavage administration according to different groups, two times/d. The indice of softtissue modeling localized damage and the concentration of IL-1β in the blood in thegroups were measured and histological changes were observed by electron microscope.Statistics software SPSS19.0was applied for data processing。Results:(1) Partially damaged soft tissue injury symptomatology index: The1st,3rd,7th,10thday after the establishment of model, the soft tissue injury symptomatologyindex in Sizhixiangfu-san group and Qili-san group were lower than that in the blankcontrol group. The symptomatology index in the three groups showed the tendency ofsteady decline. The indice in the three groups were similar in the1stand3rdday, and ithad no significant difference compared with each other (P>0.05). In the6thday, thesymptomatology index in Sizhixiangfu-san group and Qili-san group were lower thanthat in blank control group, and there was a significant difference between Qili-sangroup and blank control group (P <0.05). It had a significant difference on thesymptomatology index between Sizhixiangfu-san group and blank control group in the10thday (P <0.05). Meanwhile, there was no significant difference between Qili-sangroup and blank control group (P>0.05).(2) Histological changes (observed by electron microscope): In the1stday, thetissue edema, hyperemia, inflammatory cell infiltration and parts of the muscle fiberbreakage could be clearly seen in the three groups. In the3rdday, the tissue edema,myofibrosis, necrosis and inflammatory cell infiltration could be observed. In the6thday,little muscle fibers could be seen in the Sizhixiangfu-san and Qili-san group, and quite afew inflammatory cell infiltration could be found in the blank control group. In the10thday, there was obvious damage response in blank control group. Meanwhile, the damage syndromes in the Sizhixiangfu-san and Qili-san group were weaker than that inblank control group, such as tissue edema and inflammatory cell infiltration. It showedthat the damage syndromes in the Sizhixiangfu-san and Qili-san group were weakerthan that in blank control group, and inflammatory response was alleviated.(3) IL-1β: After the establishment of model, the concentrations of IL-1β in thethree groups kept increasing in1-3days and became highest in the3rdday, and it had asignificant difference in the Sizhixiangfu-san and Qili-san group between the1stand3rdday (P <0.05), and it had a highly significant difference in blank control group betweenthe1stand3rdday (P <0.01). Then the concentrations of IL-1β in the three groupsdecreased gradually, and in the6thday it was lower than that in the3rdday, and it had asignificant difference in the three groups (P <0.05). Furthermore, in the10thday it waslower than that in the6thday, and it had a significant difference in the three groups (P<0.05). The concentrations of IL-1β in the Sizhixiangfu-san and Qili-san group weresimilar and lower than that in blank control group in the1st,3rd,7th,10thday. It had asignificant difference on the concentrations of IL-1β in Sizhixiangfu-san group andblank control group in the10thday and in Qili-san group and blank control group inthe6th,10thday (P <0.05).Conclusion: Sizhixiangfu-san can be better in the symptoms and signs of softtissue damage in the later period of the acute soft tissue injury and restore the acute softtissue from blunt trauma. Meanwhile, it could lower the concentration of IL-1β indamaged tissue. The effects of Sizhixiangfu-san on acute soft tissue injury are possiblyrelated with reducing the concentration of inflammatory factor and relieving theinflammatory reaction.
