Objective: To explore the influence of Sizhixiangfu SAN on acute soft tissue bluntinjury in rats, through establishing acute soft tissue blunt injury model, usingSizhixiangfu SAN from famous medical expert Professor ZhengHuaiXian as a drugfor treatment. and observating Symptomatology, histology, the plasma β-endorphinsof rats.Methods: According to the table of random number method,72healthy SD ratswere put into three groups(n=24): Sizhixiangfu SAN experimental group(Agroup),Qili SAN drug control group(B group) and blank control group(Cgroup),after an acute soft tissue blunt injury by self-made impact equipment..Andthen,all groups were divided into four four phase point (n=6)randomly:1st day,3rdday,6th day,10th day, The animals were given experimental drug or physiologicalsaline,2times per day. Then, the corresponding rats weight were taken, thesymptoms of the Soft tissue injury were evaluated, the histomorphology undermicroscope was observed and the plasma β-endorphins concentration was testedunder ELISA.in each observation point. At last, the data was processed by statisticalanalysis software (SPSS19.0).Results:(1)Symptomatology observation:each group has no significant difference in1st day,3rd day(P>0.05); In6th day, The score of Qili SAN group has more significantdifference to the blank control group (P=0.01<0.05); In10th day, The score ofSizhixiangfu SAN group has significant difference to the blank control group(P=0.025<0.05).(2)Histomorphology (HE400times): In1st day, Muscle tissue of each group showededema, congestion, a large number of inflammatory cells infiltration and musclefiber fractured. In3rd day, each group showed: damaged fibers degeneration,necrosis, and inflammatory cell infiltration. In6th day, Sizhixiangfu SAN group and Qili SAN group showed a few fibrous tissue, blank control group showedinflammatory cells infiltration. In10th day, Sizhixiangfu SAN group, Qili SANgroup showed: edema muscle fiber was not obvious, inflammatory cells infiltrationless, but the blank group showed more inflammatory cells infiltration than others. Ina word, as time goes by, the edema relieve inflammatory cells infiltration reduction,It showed a tendency ofinflammatory reaction relieving. Among them, SizhixiangfuSAN group, Qili SAN group improved more significantly.(3)β-endorphins results: In1st day, there was no significant difference among eachgroup(P>0.05), In3rd day, Qili SAN group increased significantly than other groups,but there was still no significant differences. In6th day, Qili SAN group remained inhigh, blank group remained in low. But,there was still no significant difference eachother (P>0.05); In10th day, Sizhixiangfu SAN group showed a highly significantdifference,contrasting blank control group(P=0.023<0.05), and showed a significantdifference to Qili SAN group (P=0.044<0.05). But, contrasted to blank group, QiliSAN group showed no significant difference (P>0.05). In the first three observationpoints, the index of Sizhixiangfu SAN was stable; the rise was in10th day,but uponinspection, there was no significant difference(P>0.05).Qili SAN group in1st,3rd,6th,10th day, changed like "wave type", peaked in the3rd day, to the minimum in10th day,upon examination,3rd day to10th day had a significant difference(P=0.041<0.05); Blank control group in1st,3rd,6th,10th day,changed volatility,but showed a trend of fall overall, upon inspection,there was no significantdifference (P>0.05).Conclusion: Sizhixiangfu SAN and Qili SAN can reduce the symptomatology indexand Promote the repair of acute soft tissue in blunt injury both. Sizhixiangfu SANcan improve the level of β-endorphins in10th day, participate in the human body forpain control.Qili SAN can improve the level of β-endorphins in the3rd day,participate in the human body for pain control.
