OBJECTIVE: To investigate the association between HLA-DRB1*08/16alleles and familial aggregation of hepatocellular carcinoma (HCC) in highhepatocellular carcinoma incidence areas in Guangxi Zhuang AutonomousRegion.METHODS:200family members from families which have had two ormore than two patients with hepatocellular carcinoma (FHHC group) andanother200family members from families which have had none patients withany malignant tumor (FNC group) were selected and matched by same sex, age(±5years), nationality, HBsAg, living environment and living conditions as theresearch subjects in high hepatocellular carcinoma incidence areas in GuangxiZhuang Autonomous Region. Genomic DNA was extracted from peripheralblood of the research subjects,and Polymerase Chain Reaction/sequence specificprimer was used to determine the frequencies of HLA-DRB1*08/16alleles. RESULTS(:1)There was no statistical difference in the gene frequency ofHLA-DRB1*08allele between FHCC group and FNC group (6.0%vs5.5%,χ2=0.046,P=0.830);However, The frequency of HLA-DRB1*16allele in FHHCgroup was significant higher than that in FNC group (24%vs15.5%, χ2=4.559,P=0.033).(2) The frequency of HLA-DRB1*08allele did not significantlyincrease with the increase in the number of patients with HCC numbers in theFHCC group (5.4%vs6.5%,χ2=0.120,P=0.729); However,the frequency ofHLA-DRB1*16allele rose significantly with the increase in the number ofpatients with HCC numbers in the FHCC group (17.2%vs29.9%,χ2=4.401,P=0.036).(3)The frequency of HLA-DRB1*08allele in yao nationality,hannationality and zhuang nationality was that:5.7%,6.1%,5.6%;The frequencyof HLA-DRB1*16allele in those three nationality groups wasthat:21.6%,12.9%,23.9%;The result showed that There was no statisticaldifferences in the gene frequency of HLA-DRB1*08allele among yaonationality,han nationality and zhuang nationality groups (χ2=0.037, P=0.982).However,The frequency of HLA-DRB1*16allele in zhuang nationality isapparently higher than that in the han nationality(χ2=5.943, P=0.015).CONCLOSIONS:(1) There may be a significant correlation betweenHLA-DRB1*16allele and familial aggregation of hepatocellular carcinoma inhigh HCC incidence areas of Guangxi province. However, HLA-DRB1*08allele may not be associated with familial aggregation of hepatocellularcarcinoma in high HCC incidence areas of Guangxi province.(2) HLA-DRB1*08allele was a low-frequency gene in population of yaonationality,han nationality and zhuang nationality groups in high HCC incidenceareas of Guangxi; However, HLA-DRB1*16allele was a high-frequencygene;The genetic distribution of HLA-DRB1*16allele in yao,han and zhuangthree main nationalities of guangxi was not the same.(3) Further analysissuggested that HLA-DRB1*16allele may be not only the genetic characteristicsof high HCC incidence areas of Guangxi province,It may be also associatedwith familial aggregation of hepatocellular carcinoma in high HCC incidenceareas of Guangxi; However,HLA-DRB1*08allele may be just the geneticcharacteristics of high HCC incidence areas of Guangxi.
