OBJECTIVE Detecting the expression levels of cytokines interleukin-35(IL-35)? cytokines interleukin-22(IL-22)in the serum of hepatocellular carcinoma high-incidence families and carcinoma-free families,and investigating the relationship between IL-35/IL-22 and familial aggregation of hepatocellular carcinoma(HCC)in Guangxi,China.METHODSExamine the levels of cytokines IL-35 and IL-22 in the serum of 93 paired members matching from hepatocellular carcinoma high-incidence families and those from carcinoma-free families via Enzyme-linked immunosorbent as-say(ELISA).A total of 186 cases,the age range is between 2 and 78 and the average age is 28.76±10.4 in high-incidence families and those from carcinoma-free families is 27.94±10.1,there were 57 males and 36 females,75 zhuang members and 18 yao members,85 HBsAg negative members and 8 HBsAg positive members in the HCC high-incidence families and there were 57 males and 36 females,75 zhuang members and 18 yao members,85 HBsAg negative members and 8 HbsAg positive members in the carcinoma-free families.Gender ? habits ? nation ? living environment ? HBV infection ?customs-matched were required as the experimental group and the control group and their age difference within 5 years.The 93 cases of HCC high-incidence family members were divided into three groups: first(52 cases),second(14 cases),third and above(27 cases)relatives according to different genetic coefficient.As the different numbers of HCC patients in HCC high-incidence families,we divided it into two groups: 2~3 cases group(42 cases)and ?4 cases group(51 cases).The paired data were compared with Wilcoxon rank and test,and the non-paired data were compared with the independent sample mannwhitney U rank and test,and the data were reported as the median and quartile [M(P25,P75)].RESULTS(1)Compared with the members from carcinoma-free families,the serum IL-35 levels of hepatocellular carcinoma high-incidence families were higher,the overall levels were 44.801(30.142,80.113)ng/L and 36.025(28.548,48.121)ng/L,P < 0.05(Z=-3.660,P=0.000).We further analyzed the levels of IL-35 in different characteristics(including male,female,zhuang zu and yao zu,HBsAg negative,HBs Ag positive)in the experimental group members compared to control group members,and it showed that the expression levels of IL-35 were 44.811(30.432,71.941)ng/l ?43.834(29.283,92.500)ng/l ? 50.022(30.331,88.386)ng/l ?37.525(25.881,46.553)ng/l ? 48.037(32.117,82.412)ng/l ?27.439(19.901,29.147)ng/l and 36.342(29.442,46.421)ng/l ?35.155(27.983,49.189)ng/l ? 36.551(28.713,48.916)ng/l ?33.972(28.391,45.647)ng/l ? 36.997(29.840,49.600)ng/l ?27.446(21.911,29.337)ng/l,respectively.Compared with the experimental group members and the control group members,Z values were 2.202? 2.908?3.641?0.745?3.606?0.533,P values were 0.028?0.004?0.000?0.457?0.000?0.594,respectively.There were obvious significant differences between the two groups in the level of IL-35,independent from sex,age,Hbs Ag negative and zhuang zu.The serum IL-22 showed a similar overall levels in the two groups(24.683(17.769,39.319)ng/L ? 22.918(15.784,32.295)ng/L,P > 0.05,Z=-1.289,P=0.197).The levels of IL-22 in different characteristics(including male,female,zhuang zu,HBsAg negative,HBsAg positive)in two groups were 25.492(18.799,39.540)ng/l ? 24.352(16.663,36.946)ng/l ? 26.661(19.755,39.614)ng/l ? 21.300(16.170,33.564)ng/l ? 26.661(20.711,40.055)ng/l ? 12.731(8.998,13.973)ng/l and 23.727(16.041,31.597)ng/l ? 19.203(15.011,33.693)ng/l ? 23.653(16.284,32.921)ng/l?3.642(2.859,6.463)ng/l?24.395(16.739,34.061)ng/l?6.847(3.795,15.085)ng/l,respectively.Z values were 1.172?0.589?1.545 ?1.078 ?1.400,P values were 0.241 ?0.556? 0.122?0.281?0.161,respectively.No significant difference were observed in this two groups,but compared with the carcinoma-free members in HCC high-incidence families,the Yao members of serum IL-22 levels in carcinoma-free members in HCC high-incidence families were significant upregulated(Z=3.724,P= 0.000).(2)There was no significant difference in the level of IL-35 among family members with different genetic coefficient in the HCC high-incidence families group(Z=0.576,P=0.456);There was also no significant difference in the level of IL-22 among family members with different genetic coefficient in the HCC high-incidence families group(Z=1.287,P=0.288).(3)The more numbers of patients with hepatocellular carcinoma,the lower expression level of IL-35 and Il-22 in peripheral blood,but had no significant difference(Z=0.573,P=0.456),(Z=0.091,P=0.766).CONCLUSION(1)In Guangxi province,the level of IL-35 in serum may related with the incidence of familial aggregation of HCC.(2)In Guangxi province,the level of IL-21 in serum not related with the incidence of familial aggregation of HCC.(3)There was no significant correlation between the expression level of IL-35 and 22 in the serum and the different genetic coefficient of the familial aggregation of liver cancer in Guangxi.
