AIM: To explore the relationship between HLA-DRB1*03、04allele andthe familial clustering of hepatocellular carcinoma, and discover the role ofgenetic factors in the familial aggregation of liver cancer in the region of highincidence in Guangxi, China.METHODS:150members whose families have had two or even moreliver cancer patients were selected in the case group, and by the matchingmethod according the gender、enthnicity、living enviroment、living habits andage,150members whose families have had only one liver cancer case and150members from families without any cancer were included in the controls, andthen we extract the DNA from the collected peripheral blood samples to testHLA-DRB1*03、04allele by the the use of PCR-SSP.RESULTS: The expression frequency of the HLA-DRB1*03/04allele inthe case group were all higher than that of the two control groups, the differenceof the HLA-DRB1*04allele expression was significant (χ2=8.748, P=0.013).While the different distribution of the HLA-DRB1*03allele among the threegroups did not have statistical significance (χ2=1.074, P=0.584). And as for the nationalities, the HLA-DRB1*03allele had a significantly statistical differencebetween the Yao nationality and the other two nationalities(χ2=9.155,P=0.010);while the distribution of HLA-DRB1*04allele was not statisticallysignificant(χ2=2.296,P=0.317).CONLUSIONS: HLA-DRB1*04allele might be relateted to the familialaggregation of liver cancer in the high incidence area of Guangxi, China, and thecorrelation might not be because of the carriong the relevant susceptible gene ofHBV; while HLA-DRB1*03allele might not have any association with it, but itmight be one of genetic charateristics of the Yao nationality in these area. |
