Introduction Visceral fat is closely related to the pathogenesis of type2diabetes, and the fat factors released from visceral fat play an important role inoccurrence and progress of diabetes. Many studies had confirmed that theomention secreted from visceral fat was asscociated with type2diabetes and theprogress of its vascular complications.Nevertheless, the molecular geneticsresearch of omentin gene has not yet been fully developed.Objective Our study is to investigate the distribution characteristics ofomentin gene promoter single nucleotide polymorphisms(SNPs) with type2diabetes in the Southern chinese Han population, and to analyze their biologicalinformation.Methods In this study,100patients were from cohort of“the earlywarning mechanism research of impaired vascular endothelial function withtype2diabetes in the Southern chinese Han population”.These SNPs wereidentified by means of polymerase chain reaction(PCR) and DNA directsequencing technolygy. The biology information of omention gene promoter were predicated and analyzed by genetics analysis software, and the possibilityof function change about the SNPs was investigated. All of the data wereanalyzed by SPSS16.0.Results Six variants were found in whole promoter-wide of omentin genein Chinese Han diabetic population.They were-269A/G,-260A/G,-171g/a,-82a/g,-36a/g and-31g/a. Bioinformatics software AliBaba2.1analyzed that themutation loci of-269A/G、-171g/a、-36a/g、-31g/a were transcription factorbinding sites,such as Sp1, AhR, OLF1, NF-1, YY1, delta_fa, RAR-alph andREV-ErbAa,and these variants may cause the corresponding transcription factorbinding force change,resulting in a series of function change. Compared withdifferent genomes,-171loci g/g genotype have higher levels of urinary albuminexcretion rate than g/a genotype with type2diabetes,P <0.001, the differencewas statistically significant. In addition, higher serum omentin-1levels weredetected in women compared with men, P=0.013,OR=3.650,95%CI(1.309-10.176),gender is an independent factor associated with omentin-1.Conclution1.We first discover six SNP loci of omentin gene promoter regionin South China Han population with type2diabetes;2.-171loci g/g genotypehave higher levels of urinary albumin excretion rate than g/a genotype with type2diabetes, and the loci may be genetic warning site of diabetic impairedendothelial function.3. Higher serum omentin-1levels were detected in womencompared with men, gender is an independent factor associated with omentin-1.
