Objective: To study the genotype distribution of AHI1gene rs1535435andrs9494266polymorphisms in people from Suzhou,China.To explore the associationbetween the two single nucleotide polymorphisms(SNPs) with type2diabetesmellitus(T2DM) and with related clinical and metabolism.Methods:We conducted a case-control study with200patients through collectingtheir blood samples,which including100cases (T2DM with family history),100casescontrols (healthy people). In the study,height,weight,waistline,hip circumference, bloodpressure, blood glucose, lipids, insulin,C-peptide and other clinical indicators weremeasured.(2) The information of AHI1gene sequences and their corresponding sites ofthe SNP were obtained through the NCBI GenBank. Amplify the target gene, and thendetected all samples of two SNPs in the genotype with the method of RFLP improved,design fluorescent PCR primers using FAM labeled, digested by sequencing capillaryelectrophoresis genotyping.(3) calculation the frequency of two SNPs in the gene andgenotype betwween case group and control group, detect the relationship wheatherdifferent genotypes occur with T2DM, determine the clinical metabolic indicators of type2diabetes risk factors.Results (1)gender, age, diastolic blood pressure no difference in the T2DM group and NCgroup, in BMI, waist to hip ratio, systolic blood pressure, fasting glucose, triglycerides,VLDL, fasting insulin, fasting C-peptide, insulin resistance index in the two between thetwo groups,are statistical significance (P <0.05).(2)Genotype frequencies of the two samples to reach equilibrium by theHardy-Weinberg genetic equilibrium test, are a group representative. (3) rs1535435in the T2DM group, the frequency distribution of the GG genotype(85%), GA15%, no AA genotype, allele frequency of G and A were92.5%and7.5%inthe NC group, the distribution of The frequency of GG genotype88%, GA10%AA2%,the frequency of allele G and A were93%and7%. Showed no significant difference (P>0.05).(4) the frequency distribution of rs949466in the experimental group for the GGgenotype83%, GA17%, no AA genotype, the frequency of allele G and A were91.5%and8.5%.in the control group87%of the GG genotype,11%of the GA,2%of the AA.allele frequency of G and A were92.5%and7.5%. Showed no significant difference (P>0.05).(5) rs1535435and rs9494266genotype in the T2DM group and NC group were morecommon in the three genotypes GG and GA, AA type is less common, the G allele iscommon alleles, A allele for the rare allele, two polymorphisms (SNPs) is not correctedwith T2DM (P>0.05).(6) rs1535435and rs9494266genotypes of the GG, GA, the AA, is divided into theGG, GA+AA groups, between different genotypes of the clinical metabolic indicators, theT2DM group and NC group, blood pressure, weight, body mass index, fasting bloodglucose, glycosylated hemoglobin, lipids, fasting insulin, fasting C-peptide, HOMA-IR inthe different genotypes showed no significant difference not statistically significant (P>0.05).(7) rs1535435in the NC group, the weight significant differences in the differentgenotypes was statistically significant (P <0.05), as BMI, fasting glucose, glycosylatedhemoglobin not significantly different in the different genotypes, then the P value was0.0632,0.0713,0.0725, not yet see a significant difference, but close to the critical value of0.05.Conclusions1. rs1535435and rs9494266of gene frequencies and genotypefrequencies in the distribution of the T2DM group and NC group, no significant correlation may be associated with type2diabetes in Suzhou, China Han.2. In the normal population, RS1535435GA+AA genotype, and risk of type2diabetes risk may be greater than the GG genotype, but the need to continue Interrupted orlarge prospective study to be clear.
