Objectives: To study fasudil (Fas) intervention on adriamycin (ADM)-induced cardiac troponin I(cTnI), content of ATP, ADP, optical microscopy andelectron microscopy ultrastructural changes and impacts in rats, in order toinvestigate the protective effect of Fas intervention on AMD-inducedmyocardial injury.Method:32Female rats (each weights about200g) are separated into4groups randomly. Each group has8female rats: group A(normal saline group):the experimental by intraperitoneal injection normal saline, each10ml/kg, in the1,3,5,7,9,11thday(9:00AM). The14thdays beginning, each10ml/kg,6days.Group B (ADM+NS group), the experimental by intraperitoneal injectionAMD, each2.5ml/kg, in the1,3,5,7,9,11thday(9:00AM). The14thdaysbeginning, inject normal saline each10ml/kg,6days. Group C (ADM+Fas10mg/kg/day group),6times before administration the same to group B, The14thdays beginning, inject Fas each10ml/kg/day,6days. Group D(ADM+Fas2mg/kg/day group),6times before administration the same to group B. The14thdays beginning, inject Fas each2ml/kg/day,6days. In the fourth day after thelast administration, use10%chloral hydrate anesthetized rats by intraperitoneal injection. Each group respectively in Time1(before the start of theexperiment),Time2(after completion of ADM to48hours), Time3(applyFasudil after24hours after anesthesia), Time4(after anesthesia) the four timepoints collection of blood, detect serum cTnI level, content of ATP, ADP,specimens from cardiac group, observed pathological changes by lightmicroscopy, electron microscopy.Results:(1) At time2, serum cTnI level were higher significantly ingroup B, C, D when compared to group A (P <0.01), whereas there was nodifference between the B, C, D groups; moments in T3, compared to group Bthe cTnI level of C and D groups decreased significantly(P<0.01), while the C,D also showed differences between each other (P<0.01); moment in T4wecould observed obvious decline of cTnI level in group C and D when comparedto group B(P<0.01), but there was no observed difference between the twogroups (P>0.05); C, D groups had highest cTnI level at T2; The presence ofstatistical significance of group C cTnI level in T3, T4time (P<0.05) isconfirmed; there is no statistical significance in D group cTnI level in T3, T4moment (P>0.05).(2) ADP values of the four groups of rats were statisticallysignificance (P<0.05), The P value for ATP difference in C and D group is>0.05which leads insignificance in statistics, there are significant differencesbetween the other groups comparison (P<0.05).(3) The pathological score of B,C, D groups were higher compared with group A (P<0.01). But compared withgroup B, groups C, D had lower scores (P<0.05); besides there was nodifference between group C and D (P>0.05).(4) Compared with group A, theremaining three groups of myocardial ultrastructure electron microscopy were insevere change, where the most critical damage would be observed in group BAlthough group C is better than in group D, but it still can’t compare withgroup A which had clear and normal structure. Conclusion:(1) ADM would induce myocardial injury which leadssignificantly elevated in serum cTnI and decreased in myocardial ADP, ATPcontent.(2) The impacts of applying Fas intervention could be observedincludes declined in serum cTnI and increase in myocardial ADP and ATPvalues and structure of the resulting myocardial disorders would be relieved.
