Objective:To study dexmedetomidine (DEX) intervene on adriamycin (ADM) induced myocardial injury in perioperative period in rats. The aim of the research was to provide certain experiment basis in myocardial protection for cancer patients who accepted chemotherapy during perioperative anesthesia.Methods:A total of 30 female SD rats, weighing (180+/-) g, were randomly divided into three groups with 10 rats each as follows:Group A(NS), group B(ADM+NS) and group C (ADM+DEX). (1) Group A:physiological saline (NS)were intraperitoneal(IP) injection with dose 10ml/kg in the one day of the experiment, every other day, a total of 6 times, and the same dose of NS were IP injection from the beginning of the fifteenth day in all rats of group A. (2) Group B:ADM were IP injection with dose 2.5mg/kg (diluted to 10ml/kg before injection) in the one day of the experiment, every other day, a total of 6 times, and the same dose of NS with group A were IP injection from the beginning of the fourteenth day in all rats of group B. (3) Group C:ADM were IP injection with dose 2.5mg/kg (diluted to 10ml/kg before injection) in the one day of the experiment, every other day, a total of 6 times, and DEX were IP injection with dose 50ug/kg (diluted to 10ml/kg before injection) from the beginning of the fourteenth day in all rats of group C. All blood samples were collected from following time points:At T1 (experiment day 1), T2 (experiment day 3), T3 (experiment day 15), T4 (experiment day 20, and anesthetized for all rats). Myocardial tissues were detected in all anesthetized rats. The serum H-FABP, NT-pro BNP and cTnl levels were detected with ELASA method from separated serum, and myocardial mitochondrial swelling degree were detected with ELASA method too; The content of nucleotide metabolic pool (ATP, ADP and AMP) in myocardial tissues were detected with high performance liquid chromatography (HPLC), and myocardial injury pathological grading degree were observed by light microscopy.Results:(l)Serum H-FABP level:At T2 point, these in group A were lower than that in group B and group C respectively (P<0.01), but there no statistically significant difference that compared between group B and group C(P>0.05); At T3 point, there were statistically significant difference that compared between the three groups (P>0.05); At T4 point, these in group B were significantly higher than that in group A and group C respectively(P<0.01), but there no statistically significant difference that compared between group A and group C (P>0.05). (2)Serum NT-proBNP level:At T2 point, these in group A were lower than that in group B and group C respectively (P<0.01), but there no statistically significant difference that compared between group B and group C(P>0.05). At T3 point, these in group B were significantly higher than that in group A and group C respectively(P<0.05), but there no significant difference that compared between group A and group C(P>0.05); At T4 point, these in group B were significantly higher than that in group A and group C respectively (P<0.01), but no significant difference that compared between group A and group C(P>0.05). (3)Serum cTnl level:At T1 point, there were not significant difference that compared between three groups(p>0.05); At T2 point, these in group A were lower than that in group B and group C respectively (P<0.01), but no statistically significant difference that compared between group B and group C(P>0.05); At T3 point, these in group B were significantly higher than that in group A and group C respectively(P<0.05), but there no significant difference that compared between group A and group C(P>0.05); At T4 point, these in group A was statistically significant lower than that in group B(P<0.05), but there no significant difference that compared between three groups(P>0.05). (4)Mitochondrial swelling degree:These in group B were higher than that in group A and group C respectively(P>0.05), but there no significant difference that compared between the group A and group C(P>0.05). (5)Myocardial nucleotide content:ATP content were not significant difference that compared between three groups(P>0.05); ADP content in group A and group B were significant lower than that in group B respectively(P<0.05), but there no significant difference that compared between group A and group C(P>0.05); AMP and total pool of nucleotide metabolism in group B were significant lower than that in group A and group C respectively(P<0.05), but there no significant difference that compared between group A and group C(P>0.05).Conclusion:The DEX intervention in rats can reduce that serum H-FABP, NT-proBNP, cTnl levels and myocardial mitochondrial swelling degree, and can improve mitochondrial activity, alleviate edema myocardial cells in rats, and reduce myocardial tissue degeneration and necrosis; improve the content of myocardial ATP, the content of total pool of AMP and nucleotide metabolism. It was suggested that DEX intervention could provide certain protective effect on ADM induced-myocardial injury in rats.
