Objective:To study potassium channel opener nicorandil(NIC) intervene on chemotherapy drugs adriamycin (ADM) induced acute myocardial injury in perioperative period in SD rats.Methods:A total of 40 female SD rats weighing about 240-260 grams, were randomly divided into four groups, as follows:NIC low-dose group (nic-1, n=10),NIC high-dose group (nic-2, n=10), the ADM group(ADM, n=10) and the normal control group (NS, n=10). Low-dose group of NIC was administratived by intraperitoneal injection ADM (30mg/kg) in the first day of the experiment and intragastric NIC(3mg/kg) on 1,3,5,7,9,11,13 days of the experiment,20min before injection of ADM using nicorandil. The high-dose group of NIC was administratived by intraperitoneal injection ADM (30mg/kg) in the first day of the experiment and intragastric NIC(10mg/kg) on 1,3,5,7,9,11,13 days of the experiment,20min before injection of ADM using nicorandil.The ADM group was administratived by intraperitoneal injection ADM (30mg/kg) in the first day of the experiment and intragastric the same amount of normal saline as the NIC. The normal control group was administratived by intragastric the same amount of normal saline as the NIC on the 1,3,5,7,9,11,13 days of the experiment. Observe the changes in rats body weight and survival during the experiment. On day 14 the rats were anesthetized.The serum NT-pro BNP and cTnI levels were measured by extracting the eyeball blood lml. Isolated the right carotid artery 1cm,after the dright carotid artery specimens were taken, Hematoxylin-eosin staining pathological changes in the organizational structure of blood vessels was observed.Dry the heart with filter paper before weighing, calculating cardiac weight index (heart weight/body weight). Cut the left ventricular free wall myocardial tissue and place it in 10% formalin to fix and stain for pathological observation and rating of the cardiac tissue structure. The rest of the myocardial tissue was homogenized and the malondialdehyde (MDA), superoxide dismutase (SOD) content was measured.Results:(1) The rats during ADM and NIC weight before treatment did not differ significantly, both 270±10.0 g. And 14 days after making ADM treatment, compared to the control mice, nicorandil low and high dose groups the body weight of the ADM group was significantly lower (P<0.01). Compared with the control group, Nicorandil low and high dose groups showed a certain amount of weight loss (P<0.05). In the use of nicorandil treated rats, there is no significant difference between the nicorandil low and high dose groups (P>0.05).(2) Cardiac weight index (heart weight to body weight ratio) of four groups of rats at 14 days post-treatment prompt that ADM group showed a significant increase compared to the nicorandil high-dose group and normal control group (p<0.01). The ADM group and the nicorandil low dose group showed no significant difference (P> 0.05). And the Nicorandil low dose group and the normal control group, nicorandil high dose group showed no significant difference (P> 0.05).In the Nicorandil high dose group and normal control group, the difference was not statistically significant (P> 0.05).(3)The rats serum NT-proBNP levels in ADM group was higher than the normal control group and NIC high dose group (P<0.05), the serum in NIC low and high dose groups and normal control group pairwise comparisons showed no significant difference (P> 0.05).(4) cTnI levels in ADM group was significantly lower than the control group and NIC low and high dose group (P<0.05), NIC low and high dose group and normal control group pairwise comparisons showed no significant difference (P>0.05).(5) MDA levels in ADM group were significantly higher than the normal control group and NIC low and high dose group (P<0.05), NIC low and high dose group and normal control group pairwise comparisons showed no significant difference (P>0.05).(6) SOD levels in ADM group was significantly lower than the control group and NIC low and high dose group (P<0.05), NIC low and high dose group and normal control group pairwise comparisons showed no significant difference (P>0.05). (7) Microscopic observation showed that rats had significantly doxorubicin heart, vascular tissue pathology, nicorandil low and high dose group of cardiac, vascular tissue pathological changes relatively light. Cardiac pathology Rating:ADM group was significantly higher than the NS and Nic-1, Nic-2 group (P<0.05), Nic-1, Nic-2 group and NS group pairwise comparisons showed no significant difference (P> 0.05).Conclusion:(1)ADM induced-myocardial injury and carotid artery injury in rats. (2)These are indicate to provide certain protective effects on ADM induced-myocardial injury in dose 3mg, 10mg of nicorandil intervention in rats, and the protective effects of high dose of nicorandil are more obvious.
