Objective: Study preventative protection of Ginkgo biloba extract (EGb) on adriamycin (ADR)-induced myocardial injury. And to explore its mechanism. Methods: 1.32 mice divided into three experimental groups: control group;ADR-induced myocardial injury; EGb group followed by intraperitoneal injection of ADR-induced myocardial injury as a protective group. 2. serum myocardial enzymes, myocardial lipid peroxidation malondialdehyde (MDA) content and superoxide dismutase (SOD) activity changes were measured by ELISA Kit. Observation of the general change of heart. And preparation of pathology, TUNEL assay of apoptosis; bax and bcl-2 were determined by immunohistochemistry. Results: 1. ADR rats significantly increased serum myocardial enzymes. MDA content in myocardium was significantly increased, SOD activity decreased(P <0.01); EGb protection group significantly reduced the extent of enzyme increased, myocardial tissue was no significant increase in MDA content, SOD activity is not reduced. 2. ADR rats cardiomyocyte apoptosis was significantly higher than blank control group and EGb protective group (P <0.05). 3. ADR Group myocardial bax expression was significantly more than in the control group and EGb protection group (P <0.05); bcl-2 expression was significantly less than the blank control group and EGb protection group (P <0.05).Conclusion: The study shows to some extent the protective effect of Ginkgo biloba extract on doxorubicin -induced myocardial injury. Its mechanism may be related to scavenging oxygen free radicals, anti-lipid peroxidation and inhibiting cell apoptosis.
