Objective:Through the establishment of SD (Sprague wrote-Dawley) rat model of asthma, using the method of immunohistochemical detection in asthmatic rat lung tissue nuclear factor kappa B p65predominate and expression level of TLR4, discuss nuclear factor kappa B p65predominate and TLR4expression changes in asthma pathogenesis and cough and gasp the better effects on it.Methods:Fifty SD male rats,were randomized into the control group (group A),model group(group B), Kechuanning pretreated group (group D) and budesonide pretreated group (group C). The rats in group B,group D and group C were sensitized by exposure to aerosolized oval bumin to establish the model of asthma.The rats in group A were normal,rats in group D were treated by Kechuanning and rats in group D were treated by budesonide. The level of NF-κB p65and TLR4on rat lung tissue were detected by immunohistochemical staining.The counts of total leucocytes neutrophils, eosinophils (EOS), and lymphocytes in bronchoalveolar lavage(BALF) were measured by automatic whole blood count,and the pathonology changes of lung tissue were observed. Results:1.Analyze the cell numember of the bronchoalveolar lavage fluid(BALF): the total number of leukocytes,neutrophils, lymphocytes and EOS of BALF were increased in group B(P<0.05);group D compared with Group B, the total number of leukocytes, lymphocytes, EOS in BALF were decreased(P<0.05); there were not significant differences between C and D(P<0.05).2. The pathological changes in the lung tissue of rats:group A had no significant change;group B had significant inflammatory changes.Group C and group D were showed a mild inflammatory change.3. The immunohistochemical result of NF-κB p65in the tissues of rat lungs in each group:in group A,the NF-κB p65expressed weakly positive.In group B expression of NF-κB p65was significantly increased compared with group A,compared with group A,the expression of NF-κB p65was increased in group C and group D,and decreased when compared with group B.There were no significant difference in group C and group D.4.The immunohistochemical result of TLR4in the tissues of rat lungs in each group:in group A,the TLR4expressed weakly positive.In group B expression of TLR4was significantly increased compared with group A,compared with group A,the expression of TLR4was increased in group C and group D,and decreased when compared with group B.There were no significant difference in group C and group D.Conclusion:Kechuanning could reduce the inflammatory response of the lung tissue;in the lung tissue of asthmatic group TLR4,NF-κB P65expression levels were significantly increased, Kechuanning could reduce the expression of TLR4and NF-κB P65.
