1 ObjectiveIn this study,we observed the clinical efficacy of Kechuanping Granule in the treatment of asthmatic bronchitis(Cold asthma)in children,studied the effects of Kechuanping Granule on serum cytokines(IL-1?,IL-4,IL-10,IL-17,IFN-?,TNF-?)and TLR4/NF-?B signaling pathway in children with asthmatic bronchitis(Cold asthma),and explored the mechanism of Kechuanping Granule in the treatment of asthmatic bronchitis(Cold asthma),so as to provide a theory for the treatment of asthmatic bronchitis(Cold asthma)with Chinese medicine On the basis.2 MethodsThe 60 selected cases in this study were from pediatric inpatients of the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine from January 2017 to December 2018.They were randomly divided into treatment group and control group with 30 cases in each group,and another 30 healthy children as normal group.The treatment group was treated with Kechuanping Granule,while the control group was treated with Montelukast Sodium Granule.Questionnaire scale was used to record the score of clinical symptoms in children with asthmatic bronchitis(Cold asthma).ELISA was used to detect the related cytokines in peripheral blood serum of children with asthmatic bronchitis(Cold asthma).Q-PCR was used to detect the expression of TLR4/NF-?B signal pathway-related genes.Western Blot method was used to detect the expression of TLR4/NF-?B signal pathway-related proteins.SPSS 23.0 statistical software analysis compared the changes of serum related indicators before and after treatment in children with asthmatic bronchitis(Cold asthma)and the therapeutic effect.3 Results 3.1 Clinical efficacy of Kechuanping Granule in the treatment of asthmatic bronchitis(Cold asthma)By comparing the two groups,Kechuanping Granule can effectively alleviate asthma,cough,phlegm and lung wheezing in children with asthmatic bronchitis(Cold asthma),and the treatment group is better than the control group in alleviating cough,sputum and lung wheezing(P<0.01).3.2 Changes of serum cytokines in children with asthmatic bronchitis(Cold asthma)and the effect of Kechuanping Granule on themCompared with the normal group,serum IL-4,IL-10,IL-17,TNF-? and IL-1? were significantly increased in children with asthmatic bronchitis(Cold asthma)(P<0.01),and IFN-? was significantly decreased(P<0.01);IL-17,IL-4,IL-10,TNF-?,IL-1? decreased significantly after treatment(P<0.01),and IFN-? was significantly increased(P<0.01).Compared with the control group,after Kechuanping Kechuanping Granule treatment,IL-10,IL-1? decreased(P < 0.05),and IL-1? decreased significantly(P < 0.01).3.3 Changes of the expression of TLR4/NF-?B signaling pathway in children with AB and the effect of Kechuanping Granule on itCompared with the normal group,the expression of TLR4,IRAK,TRAF-6 and NF-?B mRNA in the asthmatic bronchitis(Cold asthma)group was increased(P<0.01).Compared with the normal group,the expression of TLR4 and NF-?B protein in the asthmatic bronchitis(Cold asthma)group was significantly increased(P<0.01).Compared with the control group,the expression of NF-?B mRNA in the treatment group decreased significantly(P<0.01).4 Conclusion 1)Kechuanping Granule can up-regulate anti-inflammatory factors(such as IFN-?) and down-regulate inflammatory factors(such as IL-1?,IL-4,IL-10,IL-17,TNF-?) to control inflammatory response,thereby improving airway inflammation in children with asthmatic bronchitis(Cold asthma).2)Kechuanping Granule can inhibit the phosphorylation of IRAK,regulate the activation of TLR4/NF-?B signaling pathway,improve airway inflammation,and improve the clinical symptoms of asthmatic bronchitis(Cold asthma)in children.
