Objective: To investigate the effects of RNAi silencing Survivingene on the proliferation and apoptosis of Ovarian Cancer Cell line Ov-1063.Methods: Amplification and identification of recombinant plasmid,weredivided into experimental group, blank control group, negative control group.The recombinant plasmid (pTZU6+1-shRNA-survivin), negative plasmid wastransfected into ovarian cancer Ov-1063cells, using MTT method to observethe proliferation of Ov-1063cell inhibition rate; using AO/EB staining,Annexin V/PI (AnnexinV/PI) staining and flow cytometry Ov-1063cellapoptosis; transmission electron microscope was used to observe morphologicalchanges of Ov-1063cells. Resμlts: AnnexinV/PI staining flow cytometryshowed, after transfection of24h, apoptosis of Ov-1063cells in experimentalgroup was (16.81±1.29)%, compared with the blank control group andnegative control group had significant difference (P<0.05);24h aftertransfection, cells in experimental group were visible the apoptotic bodies wereobserved by transmission electron microscope, the negative control group nosignificant change.Conclusions: transfection pTZU6+1-shRNA-survivin coμldsignificantly down-regμlate the expression of Survivin gene, and can inhibit theproliferation of Ov-1063cells, induce its apoptosis. RNAi technology hasapplied to serous cystadenocarcinoma targeting the Survivin gene suppression therapy.
