ObjectiveTo research the differential micro RNA exprsession in ovarian serous cystadenocarcinoma chemotherapy resistance and sensitive tissue,and to analyze the differential micro RNAs in term of bioinformatic technology in order to provide theory basis for studying molecular mechanism of chemotherapy resistance and reversing it.MethodsFirstly,using the microarray to search the differential micro RNAs exprsession in ovarian serous cystadenocarcinoma chemotherapy resistance and sensitive tissue;secondly,validatiing some candidate micro RNAs in expanding samples with real time PCR technology;lastly,analyzing the significant micro RNA with bioinformatic software.Results1?micro RNAs were related to ovarian serous cystadenocarcinoma chemotherapy resistance by the microarray, miR-27a-3p, miR-141-3p and miR-200b-3p were up-regulated; miR-93-3p, miR-497-5p and miR-675-3p were down-regulated.2?The expression result of miR-27a-3p, miR-93-3p,miR-141-3p,miR-200b-3p,miR-497-5p,miR-675-3p in RT-PCR was consistent with microarray.3?Up-regulated micro RNAs(miR-27a-3p, miR-141-3p and miR-200b-3p) forecast307 targeting genes,such as:GRB2?NLK?APC?ABL2?ZEB2?RNF2;down-regulated micro RNAs(miR-93-3p? miR-497-5p?miR-675-3p) forecast 53 targeting genes,such as:ALS2?KLF8?CCND2?IGF1R?ZFX?CALU.4?The result of up-regulated micro RNAs targeting genes by GO analyzing(Biological process) include positive regulation of nucleobase-containing compound metabolic process,positive regulation of macromolecule metabolic process and cell death;the result of it by KEGG Pathway analyzing include MAPK signaling pathway,TGF? signaling pathway and ErbB signaling pathway.5?SP1?TGFB2 and PAK2 take part in biological process and signaling pathway which are relation with chemotherapy resistance.Conclusion1?The microarray and bioinformatic technology could provide a new research approach for epithelial ovarian cancer chemotherapy resistance.The interrelation between micro RNAs and epithelial ovarian cancer chemotherapy resistance may provide a new thinking and strategy to reverse chemotherapy resistance.2?miRNA-27-3p and its targeting gene SP1, miRNA-141-3p and its targeting gene TGFB2, miRNA-200b-3p and its targeting gene PAK2 may play an important role in epithelial ovarian cancer chemotherapy resistance.
