Ovarian malignant tumor is one of the three biggest of femal malignant tumor,which threaten their healthy,because of its few of symptoms in the early days,later findings and earlier diversion.Hower its death rate is the first in the gynecology malignant tumors at present.Now oncogene becomes the hot spot with the development of molecular biology.PTEN is a anti-oncogene with phosphatase activity,which takes part in the cell's growth regulation,vasculogenesis and takes roles in tumor's infiltration and diversion.It has a significal roll to tumor's development.We can utilize its role of anti-oncogene through furthor study;Recant research has shown that survivin is the strongest death inhibitors gene.Due to the special character of the tissue distributing and the powerful function of resisting cell daath,survivin has become a hot spot of the studying the tumor's initial cause and cure.MMP-2 is a endocrine protease dependentde by Zn ion,which effect tumor's invasion and diversion through degrading extracellular matrix,adjusting cell's sticky,and advancing new vessels.They participate tumor's effluence, development,infiltration,diversion.,however the study is less in ovarian tumor to them,their roles and relation-ship are not very clear.This text detects their expressions in ovarian serous corpora mammillaria cystadenocarcinoma for the first time,and approachs their roles and correlation on effluence and development in ovarian carcinoms.Search early diagnosis index and flanless treatment.Objective:To investigate the expression of PTEN,survivin and MMP-2 in ovarian serous cystadeno carcinomas.To analyses their correlation with clinical stages,histological grades and provides theory supportings for invasion of ovarion carcinoma,mechanism study of infiltration and diversion and individual gene therapy,target drug treatment.Content and Methods:We detected copression of PTEN,survivin and MMP-2 by immunohistochemistry in specimens from 36 ovarian serous corpora mammillaria cystadenocarcinoma and 16 nomal ovary tissues.The samples of ovarian carcinomas include 15 cases ofⅠ-Ⅱstage and 21 cases ofⅢ-Ⅳstage;20 cases of G1-G2 grade and 16 G3 grade;13 cases of lymphaden diversion and 23 cases of without lumphaden diversion.All data were processed by SPSS version 13.0 analysis software.Result:1.The positive expression of PTEN in ovarian carcinoma(14/36,38.9%)was significantly higher than in normal ovary tissues(14/16,87.5%),benign ovarian tumors(10/12,83.3%).There was significance in difference(P<0.05),and there was no significance difference between normal ovary issues and benign ovary tumors(P>0.05).The positive expression of PTEN in gradeⅠ-Ⅱwas significantly higher than stageⅢ-Ⅳ(P<0.05),and its expression in grade G1-G2 was significantly higher than G3(P<0.05),and its expression in no lymphaden diversion was significantly higher than lymphaden diversion(P<0.05).PTEN had a negative corelation with clinal stage and tissue grade.2.The positive expression of survivin in ovarian carcinomes(28/36,77.8%)was significantly higher than nomal ovary issues(1/16,6.3%),benign ovarian tumor (3/12,25%).There was significantly different(P<0.05).The positive expression of stageⅢ-Ⅳwas significantly higher than stageⅠ-Ⅱin survivin(P<0.05).Grade G3 was significantly higher than G1-G2(P<0.05).Lymphaden diversion was significantly higher than that without diversion.(P<0.05).Survivin has a significantly positive corelation with clinical stage and tissue grade.3.The positive expression of MMP-2 in ovarian carcinomes(22/36,61.6%)was significantly higher than nomal ovary issues(2/16,12.5%),benign ovarian tumor(2/12,16.7%).There was significantly different(P<0.05).The positive expression of stageⅢ-Ⅳwas significantly higher than stageⅠ-Ⅱin MMP-2(P<0.05).Grade G3was significantly higher than G1-G2(P<0.05).Lymphadn diversion was significantly higher than that without diversion(P<0.05).MMP-2 has a significantly positive corelation with clinical stage and tissue grade.4.PTEN had a significantly negative corelation with survivin in ovarian serous corpora mammillaria cystadenocarcinoma(r_s=-0.573,p<0.05),survivin had a significantly positive corelation with MMP-2(r_s= 0.573,p<0.05).Conclusions:1.Negative expression of PTEN correlates closly with the oncogenesis of ovarian serous corpora mammillaria cystadenocarcinoma,and relates to the progression of ovarian carcinomas.It suggests that we could target on PTEN to gene therapy.2.Survivin has positive correlation with clinical stages and histological grades.Ovarian carcinoma with expression of survivin indicates high invasion ability and poor prognosis.Over expression of survivin can be observed in most of ovarian carcinoma,which shows therapeutic targeting of survivin in ovarian carcinomas is a future possibility.3.The expression of MMP-2 has positive correlation with clincal stages,histological grades and lymphaden diversion.It indicates MMP-2 participates the development of ovarian carcinomas,and a poor prognosis.4.Survivin was significantly negative correlation with PTEN,which drew out PTEN ingibited the effluence and development of ovarian cancers,and PTEN made a down-regulation to the survivin expression.Otherwise the devitalization of PTEN made a up-regulation to it.So PTEN enhanced tumor's dffluence and development.Survivin had a positive corelation with MMP-2,which drew out MMP-2 made a up-regulation to survivin and promoted the multipication,infiltration and diversion of ovarian carcinomo.The stude of PTEN,survivin,MMP-2 will provide theory according to reveal the mechanisms and development of ovarian carcinomo,and gene therapy.To detect these molecules simultaneously,we will have a better clinical evalue to estimate malignant,diversion and progn of ovarian carcers.
