Laser Scanning Confocal Biochip Detection System

The laser confocal biochip scanning (LCBS) is a new technology with developing of biochip technology, which is based on the Confocal Imaging Principle. Through the laser scanning and computer digital image manipulation, it could make the 2-D dynamic measurement of the biochip sample. LCBS system, as one support platform of the bioscience and biotechnology, is widely applied in molecular biology, basic medicine research, genic disease diagnose, clinical therapy, environmental detection and protection, biochemical weapon, etc. It is meaningful for promoting the development of modern bioscience and biotechnology. However, the market of LCBS is full of foreign products with excessively high price, it is difficult for institute especially the lab to afford even a common system. In order to improve the situation, with the help of the knowledge innovation engineering of Chinese Academy of Sciences, the LCBS with our own intellectual property rights have been researched and developed.At the beginning, this paper discussed the principle and practical probability of the system. According to the theoretical analysis, we set up a set of equipment for the system to testify the theory. We developed a sample machine by improving and perfecting the experiment equipment. The system consisted of light source, optical pumping path, information detecting path, 2-D mechanical scanning plate, auto focusing gear and control software. The LCBS debugged and measured by some users. Through the analysis of the measurement results, it is proved that the scanning speed of our system is about 201/s, the lateral resolution is about 5μm and the sensitivity is about 0.1Flu/μm2 . The capability of our system is achieved the advanced level.In my thesis, the key technologies and technical difficulty have been analyzed. Based on theories of Confocal Imaging, a design of LCBS system has been propounded and made sure the relationship of sampling frequency and optical resolution. By the requirement of biochip detection, an automatic focusing technique and a focusing evaluation function were proposed in the thesis. In the last, some matter in debugging course and the detection results of some biochip samples using the system were analyzed.
