With the development of science and technology, it is urgent for people to discover the laws of micro-world by high precision observing devices and analyzing system in such fields as Gene engineering, biomedicine, life science, material science, microelectronics and so on. In order to provide strong researching tools for the researchers in those fields to reach and exceed the world advanced level, we have developed Laser confocal scanning microscopy and Gene chip scanner. The content of this dissertation includes the following four parts:1. Discussed the optical imaging principle of laser confocal scanning microscopic imaging system whose important character is high plane resolution and high depth resolution. Analyzed the controlling technique of scanning imaging. According to the different needs for laser confocal scanning microscopy and Gene-chip scanner, two kinds of scanning methods, optics scanning and object scanning, are presented. Poir.ted out the advantage and disadvantage of the two methods. Discussed their respective; nonlinear imaging distortion, and presented the corresponding solutions.2. Based on the general algorithms of image filtering and image enhancement, presented new image filtering and image enhancement algorithm fit for the fluorescence image. According to the characteristic of fluorescence image, the analysis system for gene chip image based on template orientation and threshold segmentation is presented.3. Introduced the basic theory for the computer graphic rendering. Discussed two kinds of surface rendering methods which are equal value surface extraction and two-dimension contour. Two image mapping algorithms of the volume rendering, forward mapping and backward mapping are researched on the basis of optical model for volume rendering. Combined with the characteristic of a series of section images from laser confocal scanning microscopy, the volume buffer algorithm and maximizing algorithms are proposed in this paper.4. Developed most parts of the whole software for the laser confocal scanning microscopy system. Applied for the software invention patent and the practical patent.Through many years' hard work, we successfully developed the first commercial Laser Confocal Scanning Fluorescence Microscopy and the first Gene- chip Scanner in our country.
