Single-mode Fiber-Optical Confocal Scanning laser Microscopy (FOCSM) is a new scanning microscopic imaging system that is based on the modern optics technology and electronics technology. Duing to applying the fiber as the point source and point detector of Laser Confocal Scanning Microscopy (LCSM) to meet the confocal condition, the system can achieves excellent resolution in axial and planar direction, and can achieve 3-D imaging of microorganism with the image processing technology in computer. FOCSM is the effective method to research the microstructure in biology, which developed quickly in the applied domains of biology, medical, physical structure, and microelectronics.In this paper, the imaging theories of the single-mode fiber-optical confocal scanning laser microscopy are analyzed systemically. According to the analyses and calculating in the optical energy consuming before the PMT detecting system, the PMT and the pre-amplifier, and the electronic system that provides the functions of scanning driving, image acquisition and image transferring are designed. PC is connected to MCU by the Enhanced Parallel Port (EPP) with three software programming that is the MCU's firmware program, the kernal mode WDM progran and the user mode application program.On the basis of the former research, the hardware of FOCSM is designed and the corresponding circuit debugging working is finished. Now it can work normally and stably.
