Single-mode fiber-optical confocal scanning laser microscopy is a new scanning microscopic imaging system, which is based on the modern optics technology and electronics technology. The characteristic of point source and point detector removes the out-of-focus light that makes the image become blur. Therefore, it has excellent resolution in axial directions, and can achieve 3-D imaging of microorganism. It has a bright prospect in the applied domain of medical, biology, physical structure, and microelectronics.The imaging theory of the single-mode fiber-optical confocal scanning laser microscopy is analyzed systemically in this paper. The effect of all the key components in the whole system is also discussed theoretically. The analysis and demonstration for the whole project is completed, and the system's components and the request of the components are established. The scheme of convenient and tight planar scanning outfit is designed and developed in emphasis. The problems of scanning distortion and optical vignetting existing in the planar system are analyzed thoroughly. According to the analyses, the scanning control and image sampling electronic system, which has the functions of distortion correction and image processing, is designed.
