Single-mode fiber-optical confocal scanning laser microscopy is a new modern microscopic imaging system with high precision. It is based on confocal imaging and combined with fiber-optical technology, laser scanning technology, digital image processing technology to realize three-dimensional imaging of microorganism. It has more excellent resolution in axial direction and landscape orientation than conventional microscope. It has a bright prospect in the applied domain of biomedicine, neurology, physical structure, and microelectronics.The imaging theory of the single-mode fiber-optical confocal scanning laser microscope is analyzed systemically and amply in this paper. Several pivotal problems of system scheme are discussed emphatically. Optic coupling and Signal-to-Noise of system are analyzed in detail. The problems of optical vignetting existing in the planar system are discussed and corresponding hardware circuit with the function of distortion correction and image processing is designed. Image showing and processing software is completed and feasibility of three-dimensional image reconstruction is approved.
